    #18894226 - 09/26/13 02:13 PM (10 years, 4 months ago)

Anyone ever use ...a petri dish the size of a small plate?  i figured I would use my regular size dish to isolate a stain....then use the big one for my single and what im going to use to colonize grain with. Any suggestions

Sounds pointless to me.

You mean the size of a small dinner plate? If so, I would advise against it. When you're doing transfers you want as little of the agar medium exposed as possible. You lift the plate slightly, transfer your wedge to the new plate and close it up.

Bigger plate = more exposed agar medium = more chance of a contaminant hitting the plate. The largest plates I ever use are 100mm in diameter.

is there any use for a bigger plate? Like can i swab like three different spots and take my first isolates from that? Should I just toss this case of big dishes?

Use the sterile, disposable, and regular-sized petris that everyone here uses. If you wanna use the three-sectioned plates to clone and isolate, and then move up to the single plates for later on isolating, etc., that'll work well, too.

Cheers

I re ordered the double sectioned plates to clone and isolate. Are you saying I could use the bigger plates for the later isolating?  Im trying to find a use for these big plates instead of tossing out in the garbage

Don't do that, you're just gonna waste time and materials.

No harm in trying them if you already have them.  If you have a flow hood they should work fine.  Just work fast and don't reach over the agar.  But like you have already been told get smaller dishes.  All I use now are 60mm dishes with 4" parafilm.

thanks for that info... I just got my three compartment sterile ones in today. I guess I can put those up on a shelf for awhile now that I have these to play with. I have flow hood and am only lifting the lid for 5 secs or so. I just didnt want to throw these bigger ones in the trash . they are prepacked and sterile also. I have a case of 500 of the three compartment. That should keep me busy for awhile.
