What Temp Range is Sufficient for Long Term LC Storage?

I took a clone of a beautiful chanterelle fruit using this tek http://www.shroomery.org/forums/showflat.php/Number/6590986#6590986 and everything worked out great. I then made H2O LC from the grain spawn. They've tested clean as has each subsequent generation.
I would like to store the LCs I have made long term let's say for two to three years. I know agar is ideal but I presently don't have access to a flow hood and I don't care for using a still air box when using agar. My question is, at what temperature range can I store the LC in the refrigerator to reach the time frame of storage I have indicated? I read somewhere that RR had an eight year old H2O LC that he pulled out of the fridge and it was just as good as the day he put it away. Is 41 F a decent temperature? Will the LC die if it freezes?

I should add that I used sterilized tap water (PC'd for 30 minutes) and NOT distilled water.

After an LC is grown and tested I store in the fridge. Since no one wants to answer you. 41F is a good temp and yes it will kill it if it freezes

Thank you for responding. It looks like I am good to go then. What kind of LCs have you used and stored in your fridge and how long have they lasted? As I have stated above, I am just using plain old tap water laden with mycelium that came from a clone that was grown on rye berries. My hope is that it will survive a few years on its own without having the need to make transfers to new LC jars that contain nutrients.

why not do both if you are not confident in your lc's holding up. Many people do agar in a SAB. make some culture slants and at least you have a backup if your lc's go bad.

A SAB works great for agar work unless you're doing a ton of plates at one time. I have a flow hood and find that I use the SAB much more often than the hood.

I'm not 100% on this, but isn't what you made considered something different from a LC? I forgot what they're called but they require distilled water. The mycelium doesn't grow and enters suspended animation. You don't have any nutrients for the myc to grow on. Maybe the tap water does. Someone else can chip in.

Again, I don't think that's a LC. and I don't feel like searching what they're called. They will last indefinitely due to suspended animation.

I found the RR quote.
"I originally posted that tek over at topia in 2001 or '02. I have three jars of mycelium water in my lab refrigerator, of three separate species, that are now 8 years old and still viable.
I'm also not the only one to come up with this. Agar came up with the idea on his own without ever seeing my thread, and I believe fahtster also came up with a similar tek. Others have also I'm sure, since it's a pretty intuitive method of getting myceliated liquid.
It's far superior to generating LC from spores on sugar water, because it's at least a hundred times faster, produces much more, and clearly lasts longer. RR "
However, he doesn't declare whether he used tap water or H2O. I will do some research on suspended animation.

@ JohnnyYen and FooMan - You two have been convincing when it comes to trying out agar. I'm gonna give it a try using slants with the red vacutainer lids, vacutainers, parafilm, and anti-bacterial agar, . I will primarily use the methods in this video ( but do it in a SAB as opposed to using a flow hood (because I don't have access to one) and use much better sterile technique than the guy in the video.
Does anyone have a decent agar recipe they would like to share? Can I just mix the agar alone with water?

Frank has a great thread in his journal with a ton of different agar recipes

What rr is taking about is called a grain lc. Ive successfully stored lc's for a cpl yrs they got used up before going bad. Lc is a rly bad long term storage method. I wud rly suggest using agar and slants for storage. For doing agar in a sab I dont use extra sugar cause its more contam prone with sugar. My basic agar recipie is 7 grams of agar 7 grams of regular instant potatoe flakes to 500ml of water.
