First Time Bulk Sub - 11/22/13 UPDATE (FROM Fruiting to Contam'ing)

Hello,

On 08/31, 10 qt grain jars  were inoculated via MS. Of these 10 jars, 7 are showing growth and 1 is showing AGGRESSIVE mycelium growth. I'm talking 25% colonization vs 5% on other jars. The last 3 grain jars are not showing ANY growth.

Would it be a good idea to transfer a few grains from the aggressive mycelium jar to an agar petri and inoculating the 3 qt jars not showing any growth? All jars smell great, moisture seems to still be optimal and there are no visual signs of contams.

The goal is to ensure all these jars fully colonize within the same timeframe, as their intended use will be in a monotub with bulk sub.

Grain to agar is a good technique, however I don't think it would help in this situation to do what you're after. Why not use the fast coloniser as a master and to G2G in a bunch of new jars? Might take an extra week or so before you're ready to spawn, so unless you're pressed for time, might work better for you. Then, just use the slower ones in smaller trays/tubs etc

Before you get too carried away with preservation and expansion, I would make sure it fruits first.

Thanks for the replies, guys. I'll do just that - fast colonizing jar will be used for G2G and agar for mycelium preservation before spawning the grains to sub, in case contams take over, and for cloning/isolating after fruiting.

Update: The fast colonizing jar, as well as a few others, were shaken last night (09/19) and mycobags have been ordered. The goal is to do a G2G using this "master" jar and colonizing a few bags with 4-6qt of grain per.

Since shaking the jar last night, the mycelium in the mastas jar has started to recover just 18 hours later (my first time working with grains - perhaps this is the normal rate?). As soon as the mycobags arrive in the mail, in about 7-10 days, the preparation of grains (soak + sterilize) will begin. Expecting to spawn grains to coir/verm/gyps substrate in 21-28 days. Fingers crossed.

I took the time to make 18 agar dishes using the following concoctions:

10 dishes:
1L
9g Agar
10ml of corn syrup
5g of sweet potato flakes
1/4 tsp of gypsum
This was PC'd where one bottle was diluted when it tipped over, only used 500ml from other bottle was used.

8 dishes:
500ml
4.5g Agar
5ml of corn syrup
1/4 tsp of gypsum
~250ml of Yam water
While cooling, some of this solution clumped pretty well, and the insoluble gypsum didn't help. Solution did not yield for a full 10 dishes. Some major clumping of gypsum + solution is visible in some dishes.

Overall, making agar petris is definitely a fun and learning experience! :grin: :ooo:

How soft is your agar?  You use alot less than recommended

I touched the agar solution that I did not use, due to possible dilution, and it was a tad bit tougher than jello...

Quick question - after inoculating agar with MS, what conditions should the petri be exposed to going forward? Most agar teks do not mention the process after introducing spores or live specimens to the dishes.

Regular colonizing conditions.  Leave agar where you would leave your jars or colonizing bulks

UPDATE!!!:

On 09/30, spawn bags were G2G'd. A total of 13 quarts of colonized whole oat (dunked) grains have been spawned to pasteurized coir/verm/gyps.



2 minitubs (spawned 10/12, 10/16), 9 qt of total substrate (spawn + sub) per tub

1 monotub (10/14), 12 qt of total sub

Today, tubs were introduced to light from 5000k daylight bulb, as well as window lighting (its raining today).

This is, in fact, my very first time using grains, G2G'ing, and spawning to bulk subs. I still have roughly 10.7 quarts in 2 spawn bags, which are taking a while due to lack of air. The spawn bag was cut just above the filter patch = "no plenum of air" (per RR).

Agar work has also been completed (first time :smile:) - PE was germinated on agar petris, transferred just once (wanted to avoid cultivating a non-fruiting isolate) and agar wedge was used to inoc a single master grain jar on 10/01.

Things have really gotten interesting as of late! F%#@ yeah!

10/22 UPDATE:

I realize very few people are really watching this thread, but I'd like to update regardless =-P.

-1 minitub with 9 qt of sub was introduced to fruiting conditions on 10/18.
-A monotub, with 12 qt of sub, is now fruiting as of 10/22.
-1 minitub with 9 qt of sub is still colonizing, and
-On 10/21, I made my first pasteurized coir/verm/coffee sub and is now placed in a 50-65°F environment to try to impede contams from germinating on the coffee before full colonization.

*In summary: 2 tubs in fruiting, 2 colonizing. Total substrate amount of 42 quarts.*

The following are pics of the minitub fruited on 10/18. After fruiting, this colonized substrate was dunked with roughly 24oz of water, and nearly every ounce was absorbed. I mix my sub on the dry side to decrease colonization time.

Look at the little hyphal knots! This is my very FIRST tub, EVAR:

-10/22: dunked vs undunked AA+ grain (and a little contam growing along :tongue:)


From Left to Right:
-PE, 1st transfer -->used to inoculate PE jars illustrated below (coffee + grain jar & master grain jar)
-Original MS PE inoculation, not used for any transfers
Note: MS PE - Y, T1 stands for Multispore Penis Envy - Yam (agar), Transfer #1. 
2 agar recipes were made, one with yam (Y), one with potato (P)




All of these petris have been placed in the fridge in order to halt growth. Some petris have been in fridge for a few days, others almost a full month.

These quart jars are being colonized by PE agar wedge transfers completed in early October 2013. One grain jar will serve as a master for G2G. The other is an experimental invitro jar containing 7oz of grains, 7oz of coffee + verm. The grains were layered on TOP of the coffee until full colonization, which is when the jar was shaken up, just 2 days ago...






PS Observation on dunked vs undunked grain before spawning to sub: both grains grow at the same rate for a few days, until the 4th day, when the dunked grain outperforms undunked by a wide margin...

So let me get this right...After cutting your mycelium out of the agar you dunked it in water then transferred to new agar plate faster growth?

No he used pieces of colonized grain to inoculate his agar and he dunked some of the pieces beforettransferring to agar and didn't dunk others 

I think I got that right

Sorry for the confusion - a few changes have been made to the post...

alabamamushie is correct... the dunked vs undunked petri contains 2 colonized grains. As I was spawning this colonized grain to a monotub on 10/14, I dunked the grain in water + ice for 15 to 20min.

A single dunked grain was placed in the petri (left side) - an undunked grain was also placed in the petri (right side), for comparison.

Agar transfers were made solely for the 2 jars Penis Envy illustrated in the pics.

The dunked vs undunked "test" is from my already colonized Albino A+ grains.
