PFTek Grow - EQ Spores & TC Liquid Culture [P1n Pr0n]

Preface This is going to be my second grow after my first successful attempt which I could only flush 1.5 times due to time restraints. Before I left for my vacation, which had prevented additionals flushes, I made 4 half pint jars of liquid culture. Each jar was filled half-way and loaded with EQ & KSSS from spore syringes and Treasure Coast & Huatla from spore prints. The inoculation of the LC was done in a still air box. Before I left on my trip, there were orbs of mycelium floating in each jar. When I came home a month later, I was surprized to see the top layer of the LC covered by a thick covering of fluffy white mycelium. Upon closer inspection I thought there was a green tint on some parts of the top layer which made me think of the dreaded contam - trich. I know that it isn't trich, because it would be much more than a few greenish tinted spots after a month, but I decided to toss them (In the backyard - of course) just to be safe. When I opened the LC, there was only a slight mushroom odor... I was elated that it wasn't contamination, but since it was opened already I had to follow through. I tossed two of the four jars outside buried with some BRF and verm - One in direct sun and one under a shady tree. The next step was to start another batch of BRF cakes. I decided to compare spore prints and liquid culture in this experiment... Hope you like it! -WolfieNuke
  
Substrate Recipe: (10) ½ Pint Jars Vermiculite 4 Cups Brown Rice Flour 2 Cups Water or Dilute Coffee 2 Cups Gypsum 2 Tablespoons (Optional) Dilute Coffee Make a pot of coffee. Drink coffee. Keep the used coffee grinds and make an additional 2 cups of dilute coffee. Or, you can use 1:1 ratio of water and coffee. Brown Rice Flour Organic recommended. If organic brown rice flour is not available, buy organic brown rice and grind using a coffee/spice grinder or food processor until a flour-like consistency. Rye flour can be substituted.
Add coffee/water to vermiculite. Mix gypsum with flour. Coat the wet vermiculite with the flour/gypsum mix. Divide evenly into 10 ½ pint jars leaving ½” space at the top. Wipe dry the top of the jar, and fill the remainder of the jar with dry vermiculite. Cover self-healing lid/ring. Cover Lid with aluminum foil.
14 September, 2013 Made 10 PF Tek jars using above recipe without gypsum. Organic brown rice flour was not available, so I made brown rice flour with organic brown rice and a powerful blender/food processor. Sterilized 10 jars using PF Tek method... Steam sterilization for 90 minutes +15 for good measure. Waited for all jars to cool (approximately 5 to 6 hours)...
Inoculated: 9/14/2013 @21:00 - (4) Jars - P. Cubensis Ecuador (Spore Syringe) 9/15/2013 @02:00 - (4) Jars - P. Cubensis Treasure Coast (Liquid Culture)
Average room temperature is 67 degrees F. Will set up thermostatically controlled heater in the next few days... Until then, they'll just have to chill
21 September, 2013 - (6.5 days after inoculation) I had the heater up and running about 4 days ago, the room temp raised from 64F to 77-79F. The LC jars have exploded with mycelium growing from many points. The spore syringes are also growing, but slowly from single points where the inoculations were performed.

I was going to say, those LCs look pretty gross, but you can never actually know until you test them. Good luck.

I was definitely worried about them, but I have some faith. I think they are just overgrown. Before I left them alone for a month they were pretty clear with orbs of mycelium. Now the mycelium looks to have made a home at the top which looks similar to what an agar plate would look like if left to fully colonize with a spore solution.
Also, they have been agitated, so now the solution looks very murky.
It surprised me because I really did take full precautions when making the LC's. I also didn't have a PC so I steamed them for 90 minutes +15 for good measure.
I'll let them settle a bit and snap a few more pictures.

28 September, 2013 - (14 days after inoculation) For anyone doubting that LC's are not a good way to maximize usage of spores, here's some proof... I have only done one grow, and have done LC's from spore syringe and direct from print with no contamination. I know the LC looks terrible, but in honest truth, it's just overgrown with mycelium. The picture below is from a Treasure Coast print from a free spore ring. Yes - I said I made a LC from a spore ring print with fantastic results! The LC was made with yeast and honey. Spores were placed from print into the LC using an inoculation loop within a SAB. On top of that, the LC is about 3-4 days ahead of the spore syringes...
TC - Liquid Culture (direct from print)

A few questions for other LC users: Once you have a verified clean LC, do you just take a clean syringe with sterile water, and put clean water into the LC, and fill the syringe with LC? I can't see another way to get the LC out without sterilizing a new syringe with water and drain and fill the syringe from the LC jar. Should an LC be placed in the refrigerator for long term storage? Will the mycelium die when nutrients are all consumed, or will it go into a dormant state?
