making of a sporesyringe

Hi I made a sporeprint (of a cube) in a glass jar. I put water in the jar the same day as the print was ready. I sucked op the water in a syringe. After about two months I inoculated BRF-jars. Now after 1 week still no signs of any mycelium growth. Was my attempt from the beginning going to fail? Please excuse my poor English, I am Dutch.

As long as the water was sterile that is an acceptable way to make a spore syringe. Be patient, it can take longer than a week for growth. If you don't get any growth, look into working with agar to get a culture going.

Yea your technique is good just like your english.
Give it some more time and you should see growth.

I guess we never asked, was the cube one that you grew? If it was a wild cube, the print is probably too dirty to go straight to a syringe.

What is the temperature of the room your jars are in? My jars were at about 60-65F with no signs for quite some time... At 77-79F mycelium were seen an a matter of 3-5 days. It sounds like you need to have more patience.

You cant tell if you´ve failed or not after only one week. wait another week, if it hasn´t grown by then, your spores may have gone bad.
2 Months is a long time by the way, the spores may have germinated, but couldn´t find any nutrients to grow, so they were unable to produce enough of their own antibacterial toxins in order to make the solution "their own territory". You should have added 4% honey by weight before sterilising to enable the hyphae to grow strong in there, (and some broad-spectrum antibiotics after sterilising can also be helpful).
If your spores had air contact, they arenot sterile anymore. So you always have to assure that your myc has the best growing conditions of all the creatures in the jar. thats more important than sterility.

Btw, getting some petri dishes is the ultimate solution to any spore germination issue. I used to make it from water in which I had boiled potatoes, with a little bit of baker´s yeast and fructose or honey. And some Metacillin. An original Bert Marco Schuldes / Sam Lanceata recipe.

My jars look just fine now: lots of white spots! When fully colonised, can I crumble the cakes and cover them with a straw and dungsubstrate? What is a good way to crumble? I have read those cakes become pretty hard..

I tried a similar technique to you. My mistake was to make the syringe too quickly (I think). I believe you should leave the print for at least 12 hours so it dries a bit and will therefore mix more readily with your water. My print was probably still wet and few spores actually got into the water. Jar I nocked up with the syringe colonised incredibly slowly and I reckon his was down to too few spores in the solution
