Inoculation queries

G'day all, sorry if this is in the wrong place and feel free to move it elsewhere if that will help. I have inoculated 6 jars about 2 weeks ago which I have had in a cupboard that holds a constant 23 degrees Celsius thanks to a heat lamp and thermostat. I wet and hang a towel in the cupboard to try keep the humidity as high as possible, each day I check on things the towel has dried out so I assume it's working somewhat. My question is, only one of the jars has visible mycellium growth. Am I expecting too much to soon, should I be seeing at least some growth in all jars by now. If there is no improvement in the coming weeks could I re-inoculate the same jars or just ditch them and start again. Would I be right in assuming I could inoculate 5 new jars with the 1 good one. Any help will be greatly appreciated. Davies.

Thats no good.  2 weeks is too long all jars should have been colonizing by now.  Your best to redo them.  I have gotten away with re inoculating them, but better safe than sorry.  Sounds like there was an issue with your syringe.

Mycelium can take 3-4 weeks to show.

OP, you don't need humidity until your jars are done and fruiting.

I usually see growth within about a week :shrug: I have had jars take a little over a week before but 3 to 4 weeks  :justno:

Yea you would want to restart by the 2nd week if no growth is apparent.  Or if you have an abundance of spores you could reshoot the jars.  It seems like you have bad syringes.

I do have a few extra syringes, I might use one to give the jars another squirt. While in sterile, conditions wearing disinfected gloves and flaming the needle. Thanks for the help. Davies

As mentioned already, you don't need the humidity. Temperature is good. I keep mine around 25-30 C usually. The 3 weeks sounds a bit high. Keep them anyway if you have extra empty jars, just in case they do start to show up!

You would be surprised.  If your syringe is dilute enough, it can take awhile for compatible monokaryotic hyphae to meet.  Is it common?  No.  Is it possible, yes.  This is why shaking grain after inoculating with MS is a no-no.

It is visible but much easier to miss when you're looking for the bright, thick, white dikaryotic mycelium.

The one good jar has taken off while all others remain dormant. I just re-inoculate the 5 sleepers, if only to see what will happen for myself. Now we play the waiting game, or.... Hungry hungry hippos.
