Finally have success on agar, How much longer should I wait to do transfers?

Inoculated 10 petri dishes about 5 days ago. This looks pretty clean to me, this is what most of them look like. Does it look alright to you guys? Should I wait until the mycelium takes over the entire agar dish, or is it safe to use this noc up other petri dishes/BRF jars?

Wait until you can see outward growth that you are confident is mycelium and then take a portion as far away from the inoculation point as possible. Usually for mycology we don't streak plates like you do in a micro lab with bacteria, and only inoculate the center of the dish. so that contaminates from one section don't grow into what should be clean mycelium.

Ahh, I see. Someone told me to do it that way, but I can understand why it would be better to just do the center. I did it roughly half and half, a few of the dishes I only did one dot in the center. Thanks man.

Does the same go for taking a wedge from an isolate to put on grain? As far from inoculation point? I was under assumption you take a wedge like you would a slice of pizza from a pie.

Just a small wedge from the outer edge of growth. For 2 reasons- 1 because the edge is farthest away from possible contaminants, and more importantly because the outer edge is the fastest growing most vigorous mycelium
Edit: you also only want a small piece because the whole point of using agar is to get an isolate. You want to transfer the least amount of different genetics each time you make a transfer. A large wedge equals more possible genetics unless you already have an isolate

Does anyone else think it's mold? I used sterilized prepoured petri dishes, used a glovebox, spore syringes I bought from a reputed supplier, all of the petri dishes look consistent, and the white growth is only where the spores were injected. Is this really mold?

I usually put a nice chunk of agar in my master grain jars....like 1/5 of a plate....like a slice of pizza.

So it shouldn't matter how close to the middle at that point then? Also, is it normal for the older myc closer to the inoculation point out wards to be a little more thick and fuzzy than the leading edges?
