im curious how others maintain there cubensis genetics

i had problems with my last procedure which is as follows.

Spores >>>>  strain isolation until monocultures are established >>> monocultrues are tested on coco coir >>>
best cultures are transfered to master slants  with a piece of hard wood>>> culture slants are put into lunch bags and put in to the refridgerator for long term storage (35F).

I had 16 different isolate strains. which i worked with for 18months. however at around 14 months i noticed that my strains were no longer producing wall to wall pinsets. believing that i had run into senecense, i decided to ask on the shroomery. rodger rabbiit told me  it may not be senescense at all, that cubensis are a sub-tropical species not accustomed to temperatures a few degrees above freezing, and that this can actually cause the strain to perform differently then it had upon being freshly isolated. he told me that some of the pros on here were taking a mini fridge and adding a cooling thermostat to keep there cultures at a steady 50F.

my plans for the future are to isolate the following morphology's of cubensis:

a standard cubensis strain. it must be a super yielder

an albino cubensis. again yield is very important

a penis envy strain standard color. if i can find a strain that doesn't produce blobs that would be cool

an albino penis envy strain.

an uncut penis envy strain.

im interested in seeing if malabar coast can produce mushrooms that keep their Vail intact

also a strain that produces red spores as i will let the caps open and allow my shrooms to stain a light red color before i pick them.

my plan is to
inject PF tek jars with spores >>> clone mushrooms from the jars that look the most promising. >>> grow the cultures out on several plates until i have monocultures. and then retest via >>> agar wedge >>> grain jar >>> cococoir/vermiculite.

seriously though am i the only one that has had issues with storing long term  cubensis cultures at 35F, i had hoped to be able to go for 24 months between strain isolations.

now i feel that i will have to re-isolate re-test every few months, just so i dont end up with under performing strains. i wonder how master slants will hold up to 50F. it seems i would need to transfer to new slants more frequently as they would dry out faster, i guess ill have to try it and find out, and be cautious along the way

i need to start studying mycology so i can get a better idea of whats going on.

Raise your fridge temperature to 40F.

My strains go much longer that way since then.

I assume you are using an insulated lunchbox?

Also,

While that Johnson control unit is very nice and will work well for your application. If you're interested in saving a couple bucks try the STC-1000 temp controller. About half the price and used very often for the same purpose.

And thanks for bringing this question around. While I hadn't gotten there and actually needed to check it out, it was something that was in the back of my mind and knew it hadn't been discussed a whole lot. - at least not frequently -

thanks im going to try 40F, i felt that 50F would be to high.

yes im using an isolated lunchbox. how long are you able to keep cubensis around this way until you see decreases in fruiting performance?

thats a great find, & $21 i think ill order one.
i actually already own the johnson control unit. i wonder if the digital unit you pointed out is as reliable as my analogue though.

The STC-1000 is what I'm currently using with out any hiccups. It's a common alternative to the Johnson unit for fermentation chambers in brewing. Widely used with little issues as far as I've seen.
