I've just started some mycology for the first time and would like to know what you guys think of my jars, and I'd like some advice.

First some facts)

+- 2 weeks ago I did 12 jars of Creeper strain cubes and 12 jars of Golden Teacher cubes. I used grass seed and followed Violets wonderful RGS tek. However, it wasn't RGS, it was "Kentucky Bluegrass" seed.

Out of ALL of those jars (24) there are 8 that I'd say are getting along nicely. Two of my jars had contam early on (one was forest green, we all know what that is, and one a light/grey blue). I threw them out immediately and will be using a dry verm layer next time to try and prevent this. So 24-2 =22... 22 left after contams removed and thrown out... 22-8 (8 that I'd say are coming along nicely)... So there's 14 that are just CRAWLING. An interesting note: 7 of the 8  I'm happy with are Creeper strain and 1 is Golden Teacher. I know many people here say a cube is a cube but that's quite a coincidence.

The 8 jars that I'm happy with mostly have one "nice" side to the jar, the others are much more barren (I used 4 injection sites with the same amount of liquid from a shaken syringe)... any ideas why? Do I just need to be patient?

I was hoping, from looking at the following pictures I could get some advice:

A) My concern is that many of the jars I'm happy with are nearly all white on one side, and barely colonized whatsoever on the other (I'll include front and back photos)... Should I be worried?

B) If it's possible to tell from looking can anyone give a hint as to why the other jars (pics to follow) are moving along sluggishly.

Exhibit A (jars I feel are turning out O.K. FRONT SIDE (good side):




Exhibit B (jars I feel are turning out O.K. except for the reverse side (this one shown):





Exhibit C (some of the "sluggish" jars) ... Any advice?


Exhibit D -- A close up of some Mycelium... Because this is my first time I'd like an expert to confirm this is the goodness and not that dreaded cobweb/spider/ whatever imposter substance. (They don't smell funky, so I should be good right?)



Last but not least, Exhibit E ... I did another batch of 24 jars this time with 100% real RGS (not kentucky bluegrass)... I was concerned about them until today. When I got home from work I inspected them and found the slightest little bit of white starting on 3/24. If anyone cares the strains were Blue Meanie, Ecuador, and Brazilian. ANY suggestions anyone can give from these pics are much appreciated. Oh yea: they were inoculated on Sunday (Today being Friday). Are they just about on course?

EDIT: I forgot to mention that I screwed up and suffocated the first batch of jars for about 5 days. I put masking tape over the injection holes and had the tinfoil still on. Reading this site caused me to correct this. Does myce generally recover from this? Is there anything I can do to kick start the sluggish jars again?

Isn't it Trichoderma? ... Anywho it's not white so it's not alright :smile:

okay cool. How long does colonization usually take? I've done a couple runs and every time it takes over to about a month for the jars to become fully colonized :frown:

The first set of 24 from this post (the nice + sluggish jars) were done about two weeks ago and you can see their state. I bet it will take at least another week and probably two for the 8 nice ones to be fully colonized. I am worried about the sluggish jars, at that rate it would be 3 months LOL.

I'm new so, keep that in mind. From what I've read there are several factors to pinning:

The single most important being 100% colonization of substrate.

Other factors:

Lack of C02 buildup / O2 exposure
Light levels
the "dunk" which you should do when fruiting.

There are more I can't remember, but the thread I was reading said 100% colonization is the single most important factor.


EDIT: Oh yea, your first question: I tried once with brown rice but I didn't read much and thought I could get away with it (1/10 jars came out without contams.. I probably set some record for getting three different colours of contams in 10 jars LOL). So far, I feel the grass seed colonizes much faster than the brown rice. From what I read on this forum, rye grass seed is a very quick sub for colonization. Again though, I am new. Take this advice with a grain of salt.

Above all else I'd like some input on exhibit D please experts :smile:
Kind of worried about the fluffy appearance of this "mycelium" (possibly). I'd like to toss these jars as quick as possible if they are in fact cobweb.

It's hard to tell tbh, shake them and smell the GE port or look for rhizomorphic (ropey strands). It's probably not cobweb as that's normally thin and even a darkish gray, but it could be trich or another white mold. Tomentose mushroom mycilium and trichoderma look pretty much the same until the trich sporulates which is what the green color is.

edit: Actually after looking at it zoomed in it does look a bit cobweb like, but again it's hard to tell on visual inspection alone. I would guess that it's likely a combination of mycilium and some other white contams, but I'd at least wait for some other opinions before tossing them.

Those could have been shook a long time ago. If you would have shaken them a week ago they would be done by now.

Such a coincidence, I was just in the kitchen pounding my jars on the counter and shaking them vigorously and came back to find your post!

You don't want to shake before 20% visible colonization. This should ensure enough knock points have been distributed to finish colonization quick. I have shook jars before they were ready and it ended up taking longer than it should have.

Does the cobweb have any distinguishable smell?
I've smelled good mycelium before and I detected a FAINT wiff of that smell from that jar. So I believe there IS some actual cube myce in that jar, but if the cobweb has no smell (or the same smell) you could very well be right that it's cobweb + myce... I'm hoping it's not.

I'd really appreciate if any other experts could chime in on exhibit D.

(any rabbits around :smile: ) I bought your videos AFTER making these jars, LOL. Probably would be much better off with these jars if I had before.

Ah okay excellent.

I don't mind them taking long. This is a hobby not a source of income (I'm not even selling them if I'm successful).

As long as I didn't kill them by doing it early (I don't think they were 20% unless the center of the jars are holding a nice surprise).

Oops.

The danger is inducing pinning? Or contams? I did it in a quite clean area and did my oust / lysol routine first.

I knew opening it isn't great, but I didn't know it's the end of the line :S

Ah well, this is my first time. I figured one jar is worth getting some good input that I'm not growing something undesirable.

I will remember this lesson well.

Edit: W/rt contams: I figured as these are sitting uncovered in my apartment with 4 injection holes in them anything floating in my air would already be in there. I guess 4 little holes doesn't really compare to the whole lid being off

Yeah, generally if you open it there's no point closing it haha. No great harm in keeping em around to see what happens, but I wouldn't expect a whole lot from it.

edit: Also do you not have anything covering the holes? No micropore tape or filter disks? Normally you either use a small vermiculite layer on top ala pf-tek or you use a filter of some sort. If you're using a verm layer you're not supposed to be shaking the jars since it'll essentially remove said layer and there goes your filter.

If OP is using Violets tek he might have not realized he needs one. Violets pp5 container tek just calls for the plastic lid to be on loose for GE, but there are no holes in it as Violet does all inoculation in a SAB (as do most people these days). If he does have holes with no filters then those jars are all fucked, its only a matter of time :shrug:
