Doing agar how do you know where is best to take a transfer from?
    #18806857 - 09/06/13 10:21 AM (10 years, 5 months ago)

So I've started from spores and now on my second transfer the wedge is surrounded by rhizomorphic growth. I'd still like to do one or two more transfers to ensure a strong strain/group of strains, and there's also a small contam present on the outside of the current dish, but looking at some older threads I can't really understand what is the best place to do this. I'd take a pic, but I don't have a camera, however could anyone provide some good exampls or simply a thorough description? When looking at the past threads the descriptions didn't seem to match up with what I perceived of the accompanying pictures, but likewise the pictures were rather blurry when zooming in so I'm not sure how other posters could really tell either.

Also I had been using a sharp kitchen knife to make my transfers, but I suspect this might be a source of contaminants. Would an inoculation loop or tweezers be a better idea? The inoculation loop seems very easy to flame sterilize, but the tweezers being more dense it seems as if it's be more difficult. Can inoculation loops even be used for such work or are they generally reserved for scooping spores only?

A scalpel is what you want for transferring wedges. And have you watched RR's videos on agar transfers? They're pretty thorough in explaining where to take the transfer from. You want the most rhizomorphic part of the culture and you take it from the leading edge of the mycelium

I think I have watched his videos, but it's been awhile. So you want the piece that is growing outwards the fastest, but what exactly constitutes "most rhizomophic?" I've got some big pieces of ropy rhizomorphic growth, and then I've got pieces which I'd consider just as 'rhizomorphic' yet they're made up of a lot of smaller strands. I know what rhizomorphic growth is quite clearly, but when choosing 'the most rhizomorphic' the term becomes rather ambiguous to me, hence the confusion.

I know what you mean. I'm no expert here so id wait for someone else to chime in but personally if I had to choose one, I would choose the ropy one. If I wasn't sure what to do then I would transfer both and have multiple cultures just to be sure I didn't pick the wrong one

Stropharis' write-up on cleaning and isolation should help you out.

That was quite helpful actually, thanks for the link. So now my question is what is the next best alternative if I don't have a scalpel around? Box cutter blade maybe? They're quite thin, I'd think that would be easy enough to flame sterilize?
