Isolating a strain from grain jar *PICS*

It's a bit harder to see in the first pic, but in the 2nd pic you can clearly see some individual rhizo strands poking out a good distance above the grains. If I was to take a scalpel and very carefully slice off 1 single strand from that area and transfer it to agar, would I automatically have an isolate off the bat? And would the 1 strand have a hard time surviving alone in the Petri dish or would it be just fine?

Chances are it would grow out and sector...

Thanks for the quick reply. I assumed that 1 individual strand from the jar would be just like taking a wedge from a sector on agar but I guess not. I'm sure it would at least get me real close to an isolate though but I really don't know too much about agar so I could be wrong

It would get you close, but there would most likely still be sectors....
