How many sectors do you usually get from a clone on agar?
    #18804345 - 09/05/13 05:45 PM (10 years, 4 months ago)

I know it's gonna be different every time, but just looking for a ball park figure. Couldn't find an exact answer with the search engine

Well, it'll be different every time. I would guess 1-5. I've never really kept track of it. After you clone to agar, you've done all the hard part. Finding an iso out of a clone is very easy

Between 1-50 would be closer IMO. Sometimes I don't see sectors until I do a transfer

I have read some posts that indicate isolates from a clone may not perform as well as the original fruit.

It would appear that genes can work in concert to produce fruits, but when separated do not perform as well.

Just so you know, I'm only repeating what I've read. I have no experience in the matter. But It seems to make sense. I'll try to find some posts to back up my claims.

isolate from a clone and theres not much hassel:shrug:

Nor will there likely to be any more mushrooms.  If a fruit has multiple strains active, removing one might just ruin the whole thing since they're working in concert.

Clones often perform better as the original composites, rather than as individually isolated sectors.

I always thought that when you do agar work it's better to take a wedge so you have a bunch of sectors that aleady work well together?

I have read this as well, I just thought that after isolating a few different cultures from the clone, one of them at least would be good. But how would someone ever actually clone then and keep all the strains? Would you put a pin/clone tissue on an agar plate and then cut a huge wedge across the plate so it has all the different sectors still? And then use that to inoc a grain master which you would then use for g2g

Actually if that was done with a pin it would contaminate the grain jar. Maybe the best thing to do would be to take tissue from inside of a mature fruit and transfer it to agar to make sure it's not contaminated, then take a huge wedge from that and make an LC or grain master?

From what I've read, sectors aren't seen until a few transferes away from the MS clone. This seams reasonable if all the genetics would be growing together at first. Clone material is just dense mycelium.
