Question about test tube slants

Arent test tube slants harder to work with since they dont hav the 2-dimensional leverage of petri dishes? and thus wudnt it be almost impossible to salvage an infected mycelium from inside a slant?

Solution: don't ever put "infected" mycelium into a slant.
Petris are for dealing with contams. Slants are for long-term storage of clean cultures.

How do you clone tissue to a petri dish and then transfer to a slant without isolating a sector to keep all genetics currently present within that clone?

Cut a larger piece with multiple sectors?
You want to make a slant with multiple genetics? Why not just keep use a print, then?

I would prefer to have all the strains/genetics within the tissue that made the fruit like it is, rather than isolate a single part of it to grow it out only to find it may not fruit or it just isn't a great performer.
Im thinking if I take a small enough tissue sample and let it grow out just a tiny bit on agar, I can cut out the agar chunk small enough around the tissue and all of the growing mycellium to stick into a slant once I know there arent any contams present.

Wow i just learnt somthin new! So ur telling me even wen one clones a mushroom tissue on agar, the mycelium growing out of that tissue wont necessarily contain all the genetics of the cloned tissue?!!

Correct. You may have to transfer several times to get a mono culture, but you're way ahead compared to starting with spore print. And this way you can see the fruit you're cloning and get a hint of which traits it might have.

It will contain all the genetics, it's just that there might be multiple strains from a single fruit body.

Yes, m4b's description is accurate, it is not like more sectors will "appear" magically after transfers...they are all there from the start whether they are visibly distiniguishing themselves or not.
Slants are for long-term storage of great fruiting isolates, ones worthy of saving. There is no randomness here, nor should there be.
If you want to do MS, like pestilence said, go with spores.

So just to make sure im getting it right let me ask again, to get a mono strain/culture from the mycelia of a cloned tissue one has to keep isolating sectors? And if so how would one know wen one has attained a single strain from a multi-strained tissue?
