It needs gas exchange, Fresh air exchange doesn't come into play until fruiting.

I tape my holes during colonization. Speeds up growth imo.

Completely seal them with regular scotch or masking tape? Or do you mean with micropore?

I've found whether during initial colonization, or spawning to colonize bulk, the ability for the sub to "breathe" is imperative!
Example: I've spawned to bulk in a tinfoil tray, then sealed air tight with saran wrap; the colonization stalled completely; I then punctured holes in the wrap, covered the punctures with micropore tape and within a day, the growth exploded.
Another thing to remember - when colonizing a monotub for example; you can tape up the poly-fil holes airtight, but due to the imperfection of the manufacturing process; the lid isn't airtight, so it's still getting gas exchange.

It's is true that you have to give the substrate fresh air at some point. And it is true that there needs to be some gas exchange. But inside the jars there'll be a massive amount of CO2 build-up, this stimulates mycelium growth and prevents mushrooms from forming.
So yeah, they need air to some degree.

I agree with Pestilence also, the OP's question could be answered (correctly) in multiple ways, depending on how the reader interprets "does mycelium need fresh air?"
This post also points out the above question - what's the difference between FAE and GE? GE, I think, is either the verm layer on PF jars, or the micropore tape/polyfil on grain/quart jars. GE allows the minimal exchange of fresh air - keeping the mycelium growing, yet allowing the CO2 buildup needed; FAE - the holes in a SGFC or monotub allows for a constant flow of fresh air into the chamber required for fruiting.
That's how I look at it anyway?
