General Questions

This is my first post as a part of the Shroomery community! I just have a few general questions that may seem mundane, however I was having problems finding a straightforward answer.

1) What is the warmest temperature that Psilocybin Cubensis B+ spores inside of a spore syringe can be stored at? The reason I ask is because I recently ordered some B+ spore syringes and have inoculated a total of 12 jars (7 rye grass seed / 5 WBS). The rye grass seed has been a week with no visible mycelium growth, and the WBS has been 4 days with no visible mycelium growth. Is there a chance the spores could have been sterilized due to the outside heat while the syringes were in my mailbox or could it be something else?

2) In your opinion, what is the best Psilocybin Cubensis strain to grow using the WBS tek? I have already successfully grown B+ using the PF tek and am currently attempting to grow B+ using the WBS tek (Using Doc34's guide) and am wondering if this is the best option?

3) Although there is a lot of difference in preference from person to person; is Doc34's WBS tek the best WBS tek or is there a better alternative? If so, what changes should I make during the prep/inoculation in relation to Doc34's tek?

My apologies if these topics have already been covered in another post.

I know you aren't growing out spores from a vendor...

Your spores won't be killed in transit.

Mycelium can take a couple weeks to appear.

Cubesnis is cubensis, there is no best strain for WBS.  B+ is fine. 

Look up TranscendingLife's journal for the best way to prep WBS.  Not sure if it is different from Doc34 or not.

Click the link in my signature. It walks you through start to finish for grains.  And as mush said, it can take up to 3 weeks to see growth.

It can take up to three weeks for any type of growth to begin? Why is it that I keep reading all over that people are getting fully colonized jars using this tek within like 7-10 days? I only put 1 cc of spores in to each pint sized jar, could that be why?

1cc is plenty.  You can even use .5cc.  You could maybe have a pint of grain done in 10 days, but that is nothing but luck.

Additionally,

I am planning on using a monotub as a fruiting chamber. I am a complete novice in this regard. I have seen many people talk about "flushing" their tubs, what exactly does this mean?

A flush is when all your mushrooms come up.  Once you pick your first flush of mushrooms, you can usually get a second flush, or a third flush.

There are a lot of factors as far as why it can take longer than others.  When you deal with a Multispore Syringe(MS) you're dealing with millions of genetics.  Now, those genetics are probably a-ok, they just might be a little slow to start off, doesnt mean itll be bad.

The syringe could also be older, you have only 50% control over the storage of your syringe, the other half could be from where you got it.  Shit happens and if you approach the resource you received it from in the proper manor you will most likely get a replacement sent to you. (If it was from a vendor, remember to mention only microscopy use or be prepared to be ignored)

It could have also been inoculation method, if you didn't go down the face of the grain and between the glass it will start to colonize in the middle and you wont see growth for weeks.  If you shook the jar after inoculation you might not see any growth either.  Its almost like how humans reproduce.  The dykaryotic mycelium have to find each other and make it like mushroom bunnies (in a general sense).

Patience is all I can advise, keep them at room temp and if after 3 weeks no signs I would start over.

Yes, I shook right after inoculation. Not too much, but I have read you should in order to distribute the spores.

You shake the syringe to distribute the spores, not the jars.  You need the monokaryotic mycelium to be as close as possible.  You shake your jars at 25-50% colonization.

It'll take a while to colonize, if at all.  Most likely you will get growth, just gotta wait and see.  Worst thing that happens is that they dont find each other in some jars, shouldn't happen to all the jars unless your luck is really awful... But if you're doing grain to grain you only need 1 jar to pull through anyway.

No problem, you'll most likely see something in all your jars.  But nothing not even intended failure is 100% in this hobby.  Nature has a way.

You want the monokaryotic mycelium to be as close as possible, :wink:.  That's how dikaryotic mycelium comes about.
