Rye Grain Mycobags...

...how long should they take to colonize? I purchased 3 sterilized rye grain mycobags and inoculated each of them with about 3.5 ml of spore solution. It's been 2 weeks, and I still see no signs of mycelium growth. I also inoculated 18 BRF half pint jars at the same time with a different variety, put them in the same location, and they're about half colonized. I expected the bags to take longer, and I realize that it tends to be a few degrees warmer in the jars, but I thought I'd see a little something by now. What gives?

Actually, bags produce more heat than jars, especially 1/2 pints.
The more spawn and/or substrate colonizing, the more heat they'll produce.
Did you make sure to shake the syringe well before inoculation?
How old was the syringe?

Honestly, it sounds like a bad syringe, but wait another week.
If not growth in 3 weeks, off to the garden they go.

For some reason I thought that grain took longer to colonize. I did shake the syringe before inoculating each bag. I know there were spores in the syringe because I could see them, though I didn't prepare it myself. I inoculated the jars with two syringes of the same variety, one I made myself and one purchased, both purchased syringes were obtained at the same time. I used the syringes about a day after I got them in the mail. Though there was a bit of drama surrounding the actual shipping of the syringes and I remember thinking, "Gee, I hope these syringes are still viable after all that." They took about 14 days to arrive. I don't know how long they'd been sitting around before they were shipped to me.

If I see nothing in one more week, is there the possibility that I can prepare a syringe myself and try to re-inoculate the bags, or are they done for?

You can try to reinoculate the bags. Be sure to do it with the same variety of cube.
The myth of grains taking longer is because we're using larger containers than 1/2 pints.
Normally, we use quart jars or larger bags.
The majority of my MS inoculations take from 14-21 days for full colonization.

Be sure to allow your spores to hydrate in the syringe before using it.
A rule of thumb is to allow a syringe to sit for 5-7 days after making it to ensure the spores are hydrated.

Smell your grain tho, one of my grain bags didn't germinate. Turns out it had gone sour.

Good call. What are they supposed to smell like? They smell kind of bitter.

The inside of the bags seem wetter than necessary, but I don't know if this is supposed to happen..

They should smell like fresh mushrooms.
A sour/fermenting fruit smell is a sign of bacterial infection.

They may work with you if they were all bad, you could get more bags or jars if you approach the vendor right and mention the shroomery. If the mycelium never grew, the bags patch should smell like fresh cooked grains, almost carmely, but never sour or bitter. look at the grains, do you see little bubbles forming around them, grains look slimey? I recently lost 1 of 3 bags to the same bacterial contam from a vendor. Let them kno and try to improve your sterile tech as to rule out your own contam of product. Also its good to smell them before inoc when you first receive them

All were bad...the grains are slimy and wet. :frown: They smell like vomit. :thumbdown:

I emailed the company and asked if I could have a replacement. If they want, I'll gladly mail them the bags. :lol:

Yeah, I emailed them and hopefully they'll make it right. Though out of frustration, I went out and bought a 23 qt PC, some more canning jars, WBS, and gypsum, so I'm really not going to need the mycobags anymore. Maybe I'll see if they'll replace it with poo or something.

Something I've been made aware of time and time again with this hobby: if you want it done right, you're going to have to do it yourself. Nothing against vendors or anything, but with all the money I've spent on what turned out to be mostly unnecessary supplies, I could've easily bought a PC and done it all myself and probably saved some cash, too. Of course, there are always things you're going to need that you don't have access to. Best money I've ever spent, though, is on the Let's Grow Mushrooms videos. Man, if I would've known about those and watched them BEFORE attempting a grow...oh well, I guess you pay for your education.

If you pierced the seal of the bags I don't see how you can prove that the bags were not sterile when you received them (unless you left one untouched).  I don't think I would make the company give you a refund, but that is just me.  good luck

As far as I know, bacterial contam is highly unlikely to occur from improper inoculation procedure (we're more likely to transmit mold) and is more likely to occur with improper sterilization of the grain. It's true that it would be hard to prove, but it couldn't hurt to ask, and I might as well let them know what happened. It happened to at least one other person here from the same vendor. I wasn't an asshole about it. I'd think they'd rather know and get a chance to fix it rather than go around unknowingly selling a bad product and lose business over it.
