Salvaging a syringe
    #18777906 - 08/30/13 03:20 PM (10 years, 4 months ago)

So I got a syringe from a supplier here, a very highly trusted one too I might add, and it came with some bonus E.Coli.
Currently I'm waiting for a replacement one from them (I hope). But in the meantime I decided to try and salvage it.

I had gone on vacation about a month ago and before going I had done another set of agar slides to prove that the syringe was bad and not poor sterilization on my part. Instead of scraping the agar after I confirmed the syringe was bad, I decided to just let it keep running its course, because I noticed it started to germinate. I figured while I was gone the myc would continue to creep out beyond the bounds of where the e.coli infested water was. Because the e.coli never grew outside it seemed. So I left it in my apartment for 4 weeks near enough and when I came home, I came home to some wrecked slides. The myc was all over the place but luckily it looks like the e.coli didn't continue growing, like I figured. The slides were DRY as a bone though, and I was worried the myc might ever be dead since my apartment without AC gets to 90-100F easy in the middle of a day.

I chose 3 of the best looking clean dishes and tossed the rest and re-sterilized them (I use glass, disposable pre-sterilie plastic ones are cheap-ish but not worth it for constant culturing, where I live). Picked 7 points total across the 3 and toss them in my SAB and got to work. It seems like it worked out.

I have one question though. I didn't see a lot of rhizo growth on the 3 slides, but that may be because this is (suppose to be) Pan. Cyan and I've heard their myc is always very light and fuzzy. Regardless, I took a few slices of these clumps of white myc near the edge of the growths.

In that pic, basically right next to the one triangle cut out, that some of the kind of myc I cut out on a few of the slices.
What is that? Is that basically the formation of sclerotia?

Finally. Something funny. When I opened this slide up to take the slices, I disturbed the microclimate in there and let Oxygen in. Look what showed up when I looked today.
