15 days no growth.
    #18764425 - 08/27/13 03:16 PM (10 years, 5 months ago)

Hey guys I got some spores in a jar 15 days ago on the 12th and I still have yet to see any signs of growth at all. I was just wondering if I am doing anything wrong or what. I have been hearing that people have seen growth after 3 days. I heard that you should put them somewhere at 86 so they were for about the first week or so then after some more research apparently that was all wrong and they should be somewhere between 75-80 degrees now from my understanding. Also I heard that they should be in complete darkness then I heard that they dont need to be in complete darkness or that some sunlight is okay....so.. im not sure on that part yet really I hear two different things all the time there seems to be a lot of confusion going on about what is the best exactly. So... those are in my closet I have 2 stacks going on and one of the stacks are in that black trash bag and the others are in clear so one of the sets may recieve some sunlight from here to there. The temperature of my closet as you see is usually around 80's but sometimes at night or so can dip down to 76-78. I heard that maybe the temperature flux can slow down the growth maybe? Where I live at 2-7 is when our electric company charges a shit fucking load for electricy so we dont use our AC during those times to keep the cost down and that is when the temps can spike to sometimes 85 in my closet.

I pressure cooked the jars for an hour and started the timer when the pressure cooker started making sound. inoculated in a bathroom and I cleaned it and sprayed bleach water in the air. Idk what's going on I really hate the feeling of wasting my time.

You are correct they should be in the mid 70s. Being in the 80s wont stop the jar from colonizing, it will just make a better environment for mold to grow.

Some people like darkness, some like light. I'd say it's now accepted that a natural light cycle is beneficial like sitting in front of a window. I have done both and both have been successful.

Don't be discouraged if you don't see growth yet. Multispores can vary greatly on colonization times.

Take the foil off of them and take them out of the bags, they need air to grow.

Welcome!

Sorry to hear about the lack of growth, got two bits of advice.

1) I'd edit your original post and pull out the picture of the syringe. It has the vendors name on it.

2) Based on the name of that vendor and their reputation (search them by name here) your jars are either just really slow to start, or the syringe was bad, with the latter being a strong possibility.

when you inoc. did you go straight into the cake? or let the ms run down the side?

i once went straight in and it took a long time to see growth cuz it went from inside out.

Okay I deleted the pic thanks for the heads up, but out of curiosity why is it bad for it to show the place where I got it? Also thanks for the tip about taking the tin foil off. I figured it would help keep bacteria out of it. I ordered some more spores at The Hawks Eye, cant remember the strain at the moment which leads me to my other question. Is there really a difference in strains?

Because vendors sell spores to use to study under a microscope, not cultivate.

Different "strains" of cubes don't vary too much. Something like penis envy would be different.

I keep mine on my attic stairs. Temps range from 75 at night to 82 during the day. So far I'm on day 9 and most of my jars are close to 80 percent.

I think the dim light they receive during the day helps a ton.

Good luck man! Have some patience...easy for me to say.

Start up another set of jars.  Maybe these ones will start growing.  Maybe they won't.

I've always seen growth at the four day mark even with multispore inoculation.

Either way, if you start a new set of jars now you may have better luck.  Also, did you you inoculate towards the sides of the jars so you could see the spore solution or into the center?

I've read that bathrooms are the worst place to do an inoculation.  What was your sterile procedure like?  Did you flame the needle?

If there is contamination then they may not have ever even really started growing.

Hope it works out but I wouldn't keep my hopes up.  Start a new project that follows the 'let's grow mushrooms' videos to the letter for guaranteed results.

I didn't read the entire post but assumed your experience was what I had on the first attempt. My mistake was keeping foil on after inoculating and attempting to regulate temperature. They never produced anything. Second attempt I put 4 holes instead of 2, took the foil off, and placed them on top of my kitchen cabinets where they could get some ambient light. They are fruiting now with some LARGE/THICK stems. Very happy...

You say you pressure cooked the jars, and then inoculated them... how long did you wait to inoc? I ask because if you did it within an hour or two that would be your main problem. Can you clarify?

The first batch I put them in there probably around 45m and the second one I put over night. I had 2 syringes. Now if these jars are bad can I put the new spores that I got in the same jars or do I need to get totally new jars? Can I just cook them again?

Also I injected to the side. And is there a way to tell if they didn't start growing cause of contamination or just bad spores?

Yup.  If it's bacteria, it will stink like hell and look slimy.  If it's contaminated with mold, it will look moldy.

If you are saying that you stuck a syringe into the substrate 45mins after pressure cooking, I can almost guarantee you that first batch will be trash. The one batch left over night to cool down before innoc'ing I would give another week. As to what to do with the first batch that was not given proper cool down time - I would pressure cook the jars for 90 mins and then dump out contents, wash jars in a dishwasher or hand wash, and start over.

I disagree on this part...case in point, I just noticed my incubator has been at 85F for a good few months. Zero increased contams, noticeably faster growth over the 80F it was supposed to be.

If you have live mold spores (contams) in your jars, they are fucked. Low temps will not save that.

magnussthered, my opinion is that your syringe is bunk for whatever reason.

People say it can take 3 weeks but if you are keeping them in the 80s, you should have seen something by now.

I'd get some new spores.  Do you make your own syringes from prints?

Would you say its okay to leave jars on a windowsill??

i saw more growth in the few hours i had it there while at school but when i returned i took them down because someone told me direct sunlight is bad.
but i have it on the outside of the blinds with the blinds cracked.

i know the jars had a temperature rise because the glass facing the window was warm to touch but the glass facing the room was cool to touch.

would you say its okay to set them back up there?

I would have expected growth as long as the temperatures were satisfactory.

It could be that the spores were severely dehydrated before being placed in solution and were incapable of rehydration.

Based on being at 80F there should have been some germination.

Try again with another spore syringe.

Also if you are using Pint Jars make sure your mix colonizes quickly so the jars don't stall.

Enjoy.
