No signs of colonization across 3 rye grows. Any ideas?

After 3 failed attempts at multi-spore inoculation of rye grains, I think it's time to ask the Shroomery brain trust for some help.

Attempt #1:
Using quart jars, I followed  this grain tek. It wasn't until I got to the PC stage that I realized I needed to check how many jars I could actually fit at a time. Not good... turns out I could only fit 2 at a time, and that was laying down. I had the feeling I was royally screwing up the sterilization procedure, especially because the filter discs were probably getting wet. I inoculated anyway for experimentation, and I went on vacation for 1 week while these jars sat out. When I returned, several were clearly contaminated, and the rest showed no growth, so I the discarded them all. I was using a simple incubator in this grow (cooler with a reptile pad and thermostat to keep it @ 80 degrees).

Attempt #2:
Using half-pint jars, I followed a hybrid of the above tek with RR's "5 steps to perfect grain spawn" video. I felt better about this attempt because I was using much smaller jars and knew I could fit 10 of them in the PC. My #1 inkling of "this isn't going to work" in this grow was probably that I didn't properly dry the grains after boiling/simmering them. They seemed too wet to be loading into the jar, but I decided to be cocky and load them anyway. 7 days after inoculation, I didn't notice any growth, so I discarded it. This was also done in an incubator, but I left the cooler wide open.

Attempt #3:
Using 1-pint jars, I strictly followed RR's tek.  I  created some self-healing lids with filter disk patches for fun. I put effort into making sure the grains were properly steam dried after boiling. I also used RR's recommendation of placing the grains on toilet paper and checking for a wet spot. I did not use an incubator this time after reading RR's rants about how unnecessary they are and how room temperature is fine. I inoculated these jars on 8/19 and I still see no signs of growth. I have not yet discarded these but I'm thinking I probably should. The only potential mistake I can think of is that I had soaked the grains for ~28 hours.


I know it's a huge amount of info, but I am hoping someone can help me identify a common thread between them that I may not have considered. So far, my only thoughts have been bad syringes (they all came from the same vendor), or some kind of issue with the grains themselves. I did have a successful PF grow with syringes from the same vendor, for whatever little that's worth. I have one more syringe before I need to try to acquire some more. Thinking about just doing another PF grow to be safe.

Anyone have any suggestions? I'd be thrilled to answer any questions!

Your other jars were likely fine...  it can take up to 3 weeks for signs of growth to appear.  Learns some patience.

LOL really? For some reason I had this idea in my head that I should be able to see at least some sign of colonization after 1 week. Now I wonder if I actually saw that somewhere or if I just made it up entirely. I do think it was wise to discard the first grow, as it was actually contaminated, but now I'm wishing I had kept the second one around in a separate area just to see if anything developed.

Well anyway, that's great to know. Just in case nothing ever pops up, would you recommend using the last syringe on a PF grow, and using cultures from that grow for inoculation of more grain jars? I would assume live cultures take hold and spread much more aggressively than multispore, but I've never tried it.

If you can spare the jars leave em be for another week or two. You'd be surprised....sometimes.

I've been wanting to try rye since I moved to bulk, but I got stuck on BirdSeed and GrassSeed.

Maybe look for a tek on those, I think Frank has a good WBS prep(among other good information) or for GrassSeed Violets plastic power tek.

One should never give up after a week.  A week to ten days is about average to see the first patches of mycelium showing up.

If things don't work it out with rye for whatever reason, I will start to look into WBS. I've been avoiding it because the community seems to prefer rye on the whole. I am up for trying anything though!

Man, I have TONS of jars that don't do jack shit.  Just inoculated a whole batch of jars with Golden Teachers from one of our sponsors and that was a complete bust.  Then I switched to Cambodians and that was a bust.  I finally figured out that I just might not be putting enough solution in each jar.

It happens.

Just keep in mind that WBS preparation is a little more work to get the moisture content right. And its a little messier.

yes but more innoc points :smile:.  ive used both and wbs is easiest to find for me so i use it.

Sure, I don't expect to succeed every time, especially with how inexperienced I still am. But it would be nice to have 1 success at some point  :lol:

I just checked before going to bed... tiny amount of white fuzz appearing right under the injection port in one jar!

Assuming it's not a contam I think I am in business  :trippinbawelz:

Good last few days! Went from seeing growth at the top of one jar, to a second jar also at the top, to what looks like all 5 jars showing plenty of growth at the bottom as well. I'm getting excited, starting to feel that sense of joy again that I got from the PF grow too  :awesome:

Very glad!

I still have a ways to go though. Never built a monotub or worked with coir before. Both seem relatively easy, but (as I'm sure you all know) it's tricky trying to pick and choose different teks for each portion of a grow and figure out how to stitch it all together. I think once I have a successful grain grow from start to finish it will become a lot easier for me.
