Which variety outputs the most?
So far I have experience with KSSS and AA+.
KSSS produces way more and pins quicker than AA+.
So, which cube would produce the most from MS?

Man honesty A cube is a cube and they all rock when fruiting .They can get you massive fruits. They all are great. EQ is a great one for big fruits isolate it and get wall to wall pins and flushes.But MS always get a variety that has been around along time they have been growin and growin and tested so the good genetics are there for you to enjoy. another well test one for ms is golden teacher.

How do I isolate it?
Clone large fruits to petri dishes then throw those in grain jars?
Then, G2G? And repeat?

Well he's right man. There's a strain thread for this purpose and its state to only discuss this there. Before it gets closed ill reiterate the APE idea. Its the fattest stipes next to original PE which fruits a bit slower. No other cubensis variety I know of comes close but a second decent choice for thickness are the texas varieties. If you want "thick cubes" think "penis" as their look is what they were selected over & over for.

If you're using syringes, not sure if you are, they are a genetic crap shoot!!! You never know what you will get. IMHO
