flow hood question
    #18714922 - 08/16/13 02:22 PM (10 years, 5 months ago)

I made a flow hood using a shop fan blowing through a 99.8% HEPA filter.  It so far has worked wonderfully.  My question is am I just getting lucky or not?  I see the metal 4 inch thick ones in the videos so just wondering if what I'm using is good enough

Well, since you didn't really tell us anything about your setup, we don't know.

I bought a 99.9% HEPA air filter from Lowes.  It's the kind you put in an air conditioning unit.  The fan is a super strong shop fan.  I cant find the thing online or I'd show you

From what I'm reading it shouldn't be enough.  Weird thing is I've been really successful with it so far.

I know it looks crude but I don't have $450 for a flow hood yet.  That will be my next project.  Also its completely sealed with silicone the strap just seals it against the fan and the duct tape was there to hold it while I applied the silicone

You're just getting lucky. That can't be even close to laminar flow, which means it's blowing contams into your work space.

Search up "Schmuvbox"

How can it be blowing contams if the air is going through a 99% filter?

On laminar flow, what it is and why we need it.

There is sort of a manual, actually. The Mushroom Cultivator and Growing Gourmet And Medicinal Mushrooms (both by Paul Stamets) are both pretty damn good places to start.

Because you have turbulent flow. Turbulent flow brings in dirty air from around your workspace. Laminar flow is constant and pretty much floats contams away from your workspace before they have a chance to land. This information is easily searchable.

Still air box.

I much prefer a schmuvbox. Don't hook it into a plastic box. Use the bag.

Yeah, you're gonna be lucky to fit 10 jars in that with room to work. That's just the way it is.

Like I said, search up the Schmuvbox. People will put it down all day, but I've sworn by it for the last 4 years.

I would just use it as a still airbox if you don't have a proper flowhood.  No attached gloves or force filtered air, just two holes for your arms with some plastic sheets in front for when your not working. Any mold/bacteria will settle down and stick to the sides of your workspace.

I do my agar work inside a much smaller one (66 qt maybe). Its kind of a pain cause the ceiling isn't very high and I tend to hit the top when pouring plates.  I usually can end up fitting about six quart jars plus a master for g2g comfortably.  Which for my single AA921, I can get one load (10 qt jars) done in two SAB sessions.

Made this last night gonna start with a new spore print and new jars.

How long do you let it sit before working, and I know I can't use lysol because I'm using a butane lighter, so what else works?? bleach and water...

The idea is to moisten the inside and let contaminates settle and stick to the sides over time, then you do your work...sterilizing doesn't hurt, but it's not the main point of a SAB.

Remove the gloves. They'll pull in dirty air from around your imperfect seal. Just 2 holes in a tub is all you need.

Also skip the Lysol. Bleach water or soapy water will work. Leave the box a little wet. Give it 10-15 minutes and go to work.

Seems to me...wetting, waiting...then lifting a box, would simply allow fresh air to be drawn in along with all of the contaminates it is carrying.

If you put in some type of sealant material around the lid/base this will likely work better...

A glove box needs a good seal or it will fail do to how it works.  I have a "glove bag" myself made from a closet suit bag on a pvc frame with gloves attached and a flap over the zipper at the back.  I wipe the inside with a clorox wipe as well as everything I'm placing in it and then allow a settle time of ~ 30 miniutes before doing my work.  This has allowed me a 0% contamination rate on over 100 petri and a shit ton of jars/bags. (Apart from wild specimen that I've cloned obviously)

The whole thing wouldn't work well however without a good seal around the entire thing.  As your tub has an imperfect lid attachment however be prepared for at least a chance of contams due to that fact unless you can find a way to seal it and still have it be removable.

I used 1" thick rubber adhesive weather stripping.  trust me it's sealed.

No, that is a glove box, not a still air box.

psykonautix, detach the gloves from the box and wear them on your hands.

You do not need tyvek sleeves.

A still air box is better for this hobby than a glove box.

How are you going to flame your tools between jars...inside the box?

I already found it.
I was merely stating why I would question what someone tells me.  I'm sorry I'm not a sheep and don't want to accept the first answer I get as the right one and keep on going possibly ignorant.  I like to learn EVERYTHING I can.

UPDATE

I fixed it.

Should I just wear long sleeves until I get some tyvek sleeves in?

Nope. Just wipe your arms with alcohol.
