? from a newbie
    #1871260 - 09/01/03 03:37 PM (13 years, 3 months ago)

In one of the tek's i read quote "After the jars are steam sterilized let them cool tighten the lid and let them sit for several day's watch for any colore growths or changes in the apperance of the substrate if there are none them continue to the innoculation stage" my question is
1 is this a good technique or should I do it all in the same day
2 how long is the jar sterile until i would open it or touch it
3 would it be ok to sterilize today and innoculate tomorrow

reason being i have several jars to sterilize and only a few big pot so it is going to take hours

I would invest in a PC or you could use a boiling pot of water to steam them.

Some people have gotten good results with steaming the jars in a covered pot. Others have gotten nothing but contamination because some microorganisms can survive those temperatures. It may be worth checking for used pressure cookers at your local thrift stores if you're serious about this.

I'm also going the extra mile to sterilize my substrate in the oven and then boiling them on the stove. I would like to hear your opinion if you would share it

Adaptation-23: Lowering the Contamination Rate by Presterilization

--------------------------------------------------------------------------------
The raw ingredients of the substrate can be sterilized separately to eliminate nearly all of the contaminates. Spread the vermiculite and rice flour evenly into separate cake pans and bake in a preheated oven at 300 degrees F. for 20 minutes. While these are baking, boil a large bowl of water in for 10 or 15 minutes. Use these materials to mix the substrate. It is still necessary to sterilize the jars filled with substrate.
Advantage:
Nearly all of the possible contaminates in the raw materials will be eliminated. The sterilization of the substrate after you mix and prepare the substrate will only need to address any new contamination that you added during that process. The sterilization of the mixed substrate will be much more guaranteed. You should experience a lower contamination rate. This adaptation is particularly useful if you are not using a pressure cooker to sterilize the culture jars. Boiling the culture jars only gets them up to a temperature of 212 degrees F. This adaptation provides an opportunity to eliminate contaminates that require a higher temperature to kill them.

Disadvantages:
Extra time and effort.

I'd do it, if your not using a PC. Your chances of contam would be less!

"dont think theres any reason to use the oven before boiling. U might dry it out too much" even if i add the water after sterilizing the substrate in the oven??

If you don't mind a another newbie's opinion....

If you don't have a pressure cooker it should improve your chances of getting things sterile .. a lot! ... though you should watch the oven temp and not cook it.

On the other hand .... try doing a jar without the oven ... leave it a few days without inoculating it.
If nothing grows you were successful... if you get contaminates you know you have to go the extra mile.

Good luck!
