- Proper preparation of grain is so important to get good colonization results. - .5 per jar is ok, shouldn't be an issue. - Lesson learned, don't remove the rocker while its PC'ing
A picture says a thousand words. post a pic of those jars and one closeup.

you said your moisture was fucked up anne offered you a solid tek of both grain prep and cultivation. I advise giving it a read.

Maybe we could answer the OP's question instead of propping up a known troll like anne.

JPDancer said: So I tried my first batch of grains. WBS using Frank's tek (only soaked 12 hour). None of my 5 jars showed any growth for like 2 weeks, and i kinda stopped looking at them. Then like a week ago, I decided to look at them, and one jar had one kernel that looked colonized. This spot has expanded and looks like healthy mycelium, and another spot appeared elsewhere on the jar. Now should I assume this jar is crawling with contams or something and ditch the jar, or just assume it took this long for 2 spores to get together.
I also only used like .5 spore solution per jar, or less. Also I took my rocker off my PC just to see what would happen, so the rapid pressure shift might have fucked up my moisture in my jars, so maybe it just took so long to grow cause of weird moisture distribution?
any ideas or advice?
Don't take the rocker off the PC but this should not mess up moisture content.
Sounds like your syringe is contam'd with bacteria or just a lot of dead spores.
Most people find it difficult to really eff up the moisture content of my WBS prep, I have the same issue...it's why I love it I would either get into agar (best thing you can do in this hobby) or just get a new syringe.
I'm going to guess this thread is going off the tracks. JPD, if you have further questions about my write ups or problems you have with them, please send me a PM

JPDancer said: So one week later, I saw signs of growth on two other jars from that initial batch. I shook them all just to see what happens. One had a mycellium clump the size of a tennis ball in there, I guess I dropped the spores in the middle in haste so i never saw the growth on the outside. Well we'll see if the recover.
And Frank I inoculated two more jars with the same syringe on 8/16, wbs prepped the same way but longer soak, and PC'd and cooled properly. 1 jar showed 1 spot, the other showed 2 spots of myc growth by last night 8/22. So we'll see whats good, i'm hoping to 1 of the 2 for some G2G's. need to get on making once of those gloves boxes....
Weird. Well all you need is one good colonized jar and you can use it to inoc 10 to 15 more using g2g like you said. I don't know if you have ever done g2g but if you haven't you'll love it. It is super fast. Usually only takes around a week to 10 days! When you get your culture stock built up are you going to do a monotub?

Melting the holes works well for me. You want 1.5 or 2 inch holes. You can heat a soup can and punch it through or heat a knife and use it to cut it. I guess you could use a drill with a hole saw. I have never tried that though. Check out the teks in frankhorrigans sig.
