I'm going to keep track of agar recipes here.

I'll be trying out a whole bunch of different recipes from Aloha medicinals' cookbook.

I'm looking to use most of these for cubensis as well as gourmets. Some are species-specific though, such as the PAT. All the pictures for this thread will be cubensis cultures.

A more in-depth version of this (across multiple species) will be added to my journal at some point.

If anyone is interested feel free to join in the thread, this is going to be an on-going (and sometimes slow) process but I plan to finish it by the end of this calender year.

This is not going to be an agar tutorial initially though maybe it will become one, please refrain from asking unrelated questions.

My first goal was to measure out the agar recipes and bag them up. So here we go...

Each recipe calls for 500mL of water (tap water used unless otherwise noted).

I'm probably omitting the peptone until I have a better supplier. It is expensive and not readily available for me at this time.

0) Grain-dunk agar experiment: 10g of agar to 500mL of water used to dunk 6 quarts of colonized WBS

Here are comparison plates between isolates on grain-dunk agar and malt-extract with yeast agar. I did not use very good sterile technique so the plates are a bit spotty here or there. These are simply for a visual comparison, they are not going to grains.

Grain-dunk agar works ok, I would add some dextrose to the recipe if I try it again. Growth was pretty wispy compared to other media.

1) Malt-extract: 10g LME(a) / GLME (b), 10g agar

Pictured here is GLMEA, it works great, pretty much the standard I judge media against.

2) Malt-extract yeast: 10g ELME, 10g agar, 1g yeast

Vigorous growth. I use this regularly.

3) Malt-extract with activated carbon: 10g ELME, 10g agar, 1g activated carbon

Didn't notice much difference between standard LMEA. Not recommended.

4) Malt-extract with peptone: 10g LME, 10g agar, 1g peptone

5) Malt-extract with yeast and peptone: 10g LME, 10g agar, 1g yeast, 1g peptone

6) Malt-extract with yeast and peptone and activated carbon: 10g LME, 10g agar, 1g yeast, 1g peptone, 1g activated carbon

7) Malt-extract with millet: 10g LME, 10g agar, 10g millet flour

Growth was slower and fuzzier than I'd like. No reason to recommend.

8) 597: 10g agar, 3.75g LME, 0.3g yeast, 0.5g peptone

9) Malt-extract yeast agar with vitamins: 10g LME, 10g agar, 8g ground crickets, 1g yeast, 0.01g multivitamin

10) Malt-extract yeast agar with elements: 10g LME, 10g agar, 1g yeast, 0.3g CaMgZn

Growth was comparable to standard LMEA, not any reason to recommend adding vitamins to agar for cubes.

11) Blood worm agar: 10g agar, 5g dry blood worms

12) Krill agar: 10g agar, 10g dry krill

13) Fish flake dextrose agar: 10g agar, 3g fish flakes, 10g dextrose

This did ok. Interestingly, one of my cultures really loved the shit out of it (see above) and the rest performed better on LMEA.

14) Meal worm agar: 10g agar, 10g meal worms, 3g dextrose, 1g yeast

As you can see, cubes don't really thrive on MWA. Growth was slow and strange. I would not recommend it.

15) Cat food agar: 10g agar, 10g cat food

16) Bird seed malt agar: 10g agar, 2.5g BRF, 5g WBS, 2.5g LME, ~~0.5g peptone~~

17) Millet agar: 10g agar, 10g millet flour, optional: 1g yeast

18) Cornmeal dextrose agar: 10g agar, 25g cornmeal, 3g dextrose

19) BRF Malt-extract agar: 10g agar, 40g BRF, 2g malt, 1g yeast

20) Potato dextrose agar: 10g agar, 10g potato starch, 7g dextrose

21) PAT (cordyceps-specific): 10g agar, 100mL potato broth, 1g KH2PO4, 0.5g MgSO4, 1.5g peptone, 0.5g ammonium citrate, 15g dextrose, .025g thiamine hcl

22) Potato dextrose yeast agar: 10g agar, 10g potato starch, 7g dextrose, 1g yeast

Another de facto standard for media. This is my favorite recipe at the moment.
