Isolate Storage

I am familiar with most storage techniques, and usually store isolates in diH20. I am needing to refresh my inventory and reinoculate storage vials. I am curious if any of you use substrate as storage material.

I was thinking that maybe moist crushed corn cob, sawdust, or chopped straw might make a good storage material. Pour maybe 3-5 ml of the material, colonize it and then store it at 4 degrees. You could pull a bit off with narrow tweezers or something and incoulate plates. Has anyone tried this.

How are some the ways you store isolates?

Because I am trying to both limit the strain degenernation that occurs with storage, and increase the time interaval that I can store my cultures without growing them out and putting them back to storage, which should be done occassionally to maintain your isolates.

Please tell me more about storing isolates in diH20.
As at the moment i just use petri dishes at 4?c and
transplant every 2 or 3 months.

Slants have a relatively short "shelf life" compared to water storage which I use now. Slants are okay, I am just looking for something better.

I am also interested in filter paper storage and other solid material, if any one has thoughts and opinions on that. Filter paper storage is when you allow fungi to colonize filter paper and you store it dry (with a desicant) at room temp or at -20 deg C. We store plant pathogenic fungi like this and it takes up very little room, which is a big advantage for people with limited storage space, especially if you have a large collection. I am unsure as to the storage viability though. Fungi differ as to what they can take.
