I am doing the PF Tek for the first time (some B+ and some Koh Samui). It has gone well so far, however, it has been one month since I innoculated and I'm not seeing any pins yet. It has been over 1 week since I would call the jars 'completely colonized', with a few exceptions. A few of the jars have some 'dead spots' that are only partially colonized with mycelium (not the really thick white stuff). Also, a couple jars got too hot during sterilization and there is a very small area of cooked grain at the bottom, I believe. The majority of the jars are completely covered with thick white mycelium and have been for some time. I mixed the substrate in PF's maximum fruiting formula - in this ratio:
1/4 cup BRF
1/2 cup verm
60 mLs H20

How much light (from a small regular lamp) should I give them to trigger pinning? When is it OK to force them by cold shocking? I'd say my room is about 70-75 F on average. I'm doing cakes for now, not casing.

Well, if you were patient enough to read this, thanks for any help you can give a beginner!

Just be patient and let them colonize 100%. Are you looking for pins invitro? They don't always happen. If you don't see any 2 wks after they get birthed then maybe worry. Patients is the name of the game.

Dunk and cold shock them now for 24 hours

75 degrees is optimal for fruiting

Thanks for your help so far!

Can someone point me to info about dunking and shocking? I can't seem to find it in the FAQ. I thought dunking was only for after the first flush. Isn't it risky to birth before you see in vitro pins - PFTek says fruiting is relative to in vitro pin #s.

ok dude, personally I would birth about 3-5 days after 100% colonization, I assume your using a perllite terrarium, so open your jars, fork out the top verm layer, birth the cake into a plate covered in verm, roll it around till the cake is covered, then set in your terrarium, after that you will apply the double ended casing tech, simply spoon a thin layer of moist verm onto the top of the cakes. Then "natural light cycle works great" but if you insist on using a lamp you can give them as much light as you want untill you see pins "about 7 days" just make sure they get at least 4 hrs of light untill you get pins. I think its better not to dunk untill after first flush, but all youll do is put them in a jar filled with water, in the fridge for 24 hrs......... anything else?

wtf,

dont' wait so long. 2-3 days after 100% colonized you should dunk and then birth.  Don't worry about getting them to pin in the jars.

i dont scoop the dry vermiculite >..keeping it on >raises the probability for hyphal knots..which im gettin on one cake ...they grown to spikey stipes..about to pin i think..while the other are just occuring with patches of pin heads...each dunkes 18 hours,and rolled ...with verm....

Grow Log
Tek : PF
Strain : South American [SA]
Spores : Came from homemade prints. 36 total jars to be injected.
Substrate :Brown Rice Flour [BRF] and Vermiculite [Verm]

10:15 PM 3/4/03 : Project is started. Jar prep is just putting small holes in the lids for the spore syringe and then taping them closed until inoculation. Tape used is the standard black electrical tape. First batch of jars is mixed. The mix used is 2 parts verm, 1 part water, and 1 part brf. [ This mix is the max fruiting formula by PF.] Mix is put into 1/2 pint canning jars lightly, not packed in, and left it airy. Put substrate in until it reaches the threads of the jar, then add dry verm the rest of the way.
Jars are PCd at 10psi or 240f for a period of 45 minutes. Then jars were left to sit overnight to cool.



8:39 PM 3/5/2003 : First batch of jars injected. (24 jars)

10:04 AM 3/6/2003 : Incubator is running to low 77F. Going to turn it up today.



5:45 PM 3/6/2003 : Second batch of jars is in the PC will be injected tomorrow. Turned up incubator runs at 83F. 86F is the best and if the incubator is 83F and myc growing in the jars is suppose to create a little heat I figure this should work out well.

1:11 PM 3/7/2003 : Second batch of jars is done. (12 jars)

12:10 PM 3/9/2003 : Jars are doing good, no signs of contams



2:54 AM 3/19/2003 : Tape removed from the holes today and coffee filters put over the top of the jars for added protection. Holes were opened so the CO2 in the jars could escape. I gave 12 jars to a friend who kept fucking up. 24 jar?s about 75%done.



12:13 PM 3/22/2003 : Well it's Saturday and the jars will probably be done by the 25th. So it was 36 jars. 12 were given to a friend, 24 done by Tuesday.

10:26 PM 3/23/2003 : 15 jars were done enough to put into dunk. I dunk and cold shock before birthing and after every flush. The other 9 jars have a spot the size of a pea that has to be covered so I'll do those on the 25th.



7:17 PM 3/24/2003 : 15 jars put into the pod. Put in and put a layer of moist verm on top. With geolite I just put the cakes on it. With some other chambers I put the cake on a lid filled with verm. This is done to give the shrooms on the bottom and the top of the cake some added moisture.
The other 9 jars just finished so I'm going to give them a couple more days to make sure the middle of the cake is done.



4:57 PM 3/27/2003 : 5 More cakes birthed today, 4 left going to do them tomorrow or 3/29/2003

3:58 PM 3/29/2003 : Well I lost another cake. My wife's little 1 cake pod made from a 2 liter and a small fish pump and a little geolite used up her last cake so I gave her a SA to grow. The last 3 are in the cold shock dunk.
So the total is 36 jars done all colonized with no contams. 12 given to a friend, 1 to the wife, 23 mine

12:08 PM 3/30/2003 : Last of jars put in

12:02 PM 3/31/2003 : The first 15 cakes are pinning nicely and some have shrooms coming from the bottom.



11:01 AM 4/3/2003 : Things look to be going well.



1:02 AM 4/4/2003 : Took the first flush from four of the cakes. More will be done later today.



5/8/2003 : Total 23 cakes 164 grams dried.

Update:

I cold-shocked the jars and birthed them 8 days ago. Still no sign of pins, although one cake has some green mold, so I'm throwing that one away. I'm using a simple perlite terrarium. The temp is around 75 and I've been leaving a regular incandescent lamp on them for most of the day. The humidity should be OK; I can see moisture condensing on the sides of the terrarium. Any other suggestions to get these things started? At what point will I know that nothing is going to happen with these cakes? I'm ready to birth a second batch and don't want to make the same mistake twice. Thanks for your help

Toss the bad one obviously, and mist the rest with some H20/H2O2. As far as pins go, just wait another week or so.

Don't feel so bad mindfunk, I had 10 BRF jars of Golden Teacher and only 1 pinned really good another cake has just 1 pin on it and the other 8 are still sitting in my terrarium in the small hopes that I'll wake up tomarrow to a pleasant surprise.

Thanks for the advice, folks. How can I avoid getting the green mold on my cakes? I have to open the terrarium a few times a day to fan it, and that air obviously isn't sterile. I put some dry verm on top of the cakes, but I didn't roll them in it.

OK, hopefully this batch will perk up and get rolling...in the meantime, my next set of jars are ready to be birthed. I'm thinking of switching to a cool mist humidifier, as it seems like the simplest effective method. People are saying in their teks that they cycle the cool mist on and off. Is this because there is too much fanning or too much humidity? Is there such a thing as too much fanning? It seems like my contamination problems would have been avoided if I didn't have to open the Rubbermaid so often to manually fan the air out. Also, I should cover all inlets and outlets with HEPA filters, correct? Any more advice?

Add H2O2, hydrogen peroxide, to perlite. I cant tell you how much because I dont know the size of your terrarium. But, just pour a little bit in. This will lengthen the sterility time. You need to increase humidity and expose to blue spectrum flourescent light for 12 hrs a day. Use a cool mist humidifier for 15 min every three hours. I used the same humidification method that you have when I first got started and I got nothing. So I switched to ultrasonic and that did the trick, but then I began to see green mold inside my terrarium from to much moisture so I went to cool mist.

Also you might want to case the cakes, not like pf, that never worked for me. Take a small tin foil pan, make sure you clean it with soap and water or steam it, put a layer of soaked vermiculite in bottom, slice the cakes into four sections, lay them in the tray then cover with soaked verm. Pressure cook the vermiculite the same way as the cakes before innoculation. This is to completely sterilize the vermiculite. The trick to pinning is to expose as much of the mycelia to water as possible while at the same time giving a footing for the mushrooms to grow from. Use disstilled water and pressure cook the verm. befor exposure to mycelia.
