So I've just been having fun learning agar/SAB work and watching stuff grow on it. Thought I'd share my observations and also use this thread for my G. tsugae clone grow.
Here's the original petri with the piece of tissue on it. It was actually perfectly clean from the start until I opened the lid near the top of my SAB to take a picture of the growth, then the yeast (is that yeast? Or something else?) showed up. Transferred away just fine and have a new clean dish so no worries, I've just been watching this one for fun.

It was strange, the growth was thick and extremely slow, until right when it ran into those two big yeast colonies. Then it jumped across the agar at record speed, all thin and wispy though, and started attacking the other yeast colonies. At least that's what it looks like to me, although I have no idea what I'm talking about when it comes to agar Just thought it was interesting.
Anyway, like I said it was really slow on agar but seemed to pick up speed when I inoculated rye with it. Here's the jar I started with 4 wedges:

Looking pretty nice so far, hopefully I can get it to fruit!

Those are bacteria colonies. I would suspect the thin wispy myc is some other contam also. Where you pulled your samples from is what I would call typical tsugae growth.

Thanks Jeff, good to know. I hadn't thought of that but it makes sense... Oh well it's all down the drain now anyway.
The transfer still looks nice and clean but I think I'll refrain from trying to take pictures of it...

Any idea on the mystery contam? ...cobweb? I can spot bacteria a mile away in a jar but I'm new to "contams on agar", lol

Couls be that random yet-to sporulate green mold I always find on G. tsugae. I've had really bad luck trying to clone this between that and the bacteria that seems to naturally live on, & in this species locally. Tried again last night tho with antibacterial agar and peroxide tissue dip. My faith is on the low scale but hangin' on!

my try looked a lot like this though, but really like yellow reishi 'skin', with that shiny look. and it crawled slowly. but a few were contam free, not sure how but various contams eventually penetrated and won.

Hard to say what the other is. Could be random mush spores or some type of mold.

seemed to be a phenotypic characteristic to me, but I guess it could have been a contam. Thing is, it was white at the leading edge, and as it grew and aged the older myc turned into that 'skin' or 'armor' that you see on blocks some times. Or what reishi fruit skin looks like. After about two weeks the clone grew to about a 1/2 dollar size with about 1/2 or 2/3 of it being yellowish.

That's exactly what's happening to mine. Even the clean dish. Thought it was strange...

Well, the jar is colonized... weirdly. This is the one spawned from the same dish the contaminant showed up in, so based on that and the way it looks I'd say it's probably going to fail. But I've got nothing to lose but 3 cups of pellets so I'm going to spawn it anyway.

I wonder if that strange line is the line between mycelium vs. contaminant... 
Going to start two new jars from the clean dish tomorrow, I'm not giving up yet!

Update:
The weirdly colonized jar was spawned and colonized quickly but developed a sour smell and just didn't seem right, so it got tossed. Kinda what I expected.
Started 2 new jars, one looked like the first one but the other:

Actually looks like it should. Man this stuff gets thick. Had that nice mushroomy smell at spawning, no off smells, so I think there's a good chance this one is not infected. We'll see how the block comes along!

Well, we've got a colonized bag. Clean as it gets 

Spawned on 7-31, actually only took about 5 days to completely colonize, then got REAL thick and white.
So now, anyone know how to make him fruit?

Heat, light, and time!
I wanted to run this on hemlock substrate sometime. Did you get any of this out there? I bet the mycelium from that block would run easily

You can wait for them to pin in-vitro and grow out antlers or cut small x's in the side and do conks.

I've still got the culture on a petri, but I haven't decided if I want to waste some of my last few pre-poured dishes cleaning it up and saving it, if I can't get it to fruit anyway... Although I did really want to get some dowels going and inoculate some dead hemlocks before fall. It is an interesting culture, slow as hell on agar but once I finally got a clean grain jar it colonized like mad, and I couldn't believe how quick the bag was done!
I've really got to give this agar pouring thing a go, just haven't figured out the best way and got around to it yet! Then I can mess around more.
Jeff - yeah that was my plan, just like regular reishi. I just haven't seen a lot of grows of tsugae and wasn't sure if it needed some specific pinning trigger or some kind of forest soil casing layer or anything...
For now I'm not cutting anything till I see primordia or something!

Interesting, Forester. I can grow the hell out of g. lucidum, but have had zero luck with g. curtisii. Colonizes like a mad thing, then sits there and does nothing. Zero. It sounds like tsugae may be pretty much like that. Maybe that's why we don't see any grows of it on the interwebs!! Admire your perseverance!

Yeah, that's what I was afraid of. I did see this thread from a few years ago, but all the pics are missing so I can't really tell for sure if he fruited it or not.
I figure I'll let it sit over the winter and if it doesn't fruit by spring maybe I'll throw some forest soil and moss on top of it.
