How Do LC's Work?

I have a pretty simple question, I think: if I take a multispore syringe of any arbitrary cubensis strain, and I make an LC out of it, and then I inoculate a jar of rye, and then that jar colonizes well, does that mean I can just knock-up a bunch of other jars, spawn to bulk, fruit, and then expect good results?

i wouldn't say good results, you would still be starting from multi-spore which is never the best route for any lc because it was likely fruited in open air. but if you get it to work your results would still be a multi spore guestimate

I guess the only thing you would be doing is allowing the spores to germinate in the LC and increasing the amount of fluid you can use to inoculate and maximize how many jars you can inoculate with a spore syringe. The results wouldn't be any different than using a spore syringe, minus the germination time (3-5 days).

So, assuming you've successfully colonized dozens and dozens of jars, and also had a handful of jars contaminate, you stand a chance of being able to determine whether or not your test jar is successfully colonized with nothing but your desired mycelium.  If on the other hand, you're not that familiar with what a contaminated versus clean grain jar looks like, you're going to need to spawn and fruit your test jar before you can be sure it was successful.  That's going to take a while.  What's your LC going to be doing in the meantime?  Are you prepared to wait a month or more in order to test your LC before deploying it?

I know agar work seems intimidating, especially because a lot of people focus on isolating strains.  However, learning to use agar to ensure a clean sample, and/or transfer away from any contaminants that show up, is dead easy.  You can do it in a still air box using widemouth half-pints as petri dishes.  Then you can use a wedge to inoculate a master grain jar, then G2G that jar to 6 more, then those 6 to 36 more, until the sheer mathematics of the thing results in you not having anywhere to sit down because your entire house is full of jars full of mycelium.

Grain to grain transfers are easy to do, and have a very low contamination rate.  They also don't require any syringes, or injection ports, or magnetic stir bars.  Just jars and filter lids.

Don't believe the hype.

Well I was meaning that now I have a colonized jar of rye from an LC. It is white and fluffy. It looks great. Can I use the same LC to make more jars, and should they fruit just fine? Alternatively, can I use the colonized jar to do G2G? In other words, if I make an LC and it colonizes a jar, does that automatically mean that it's a good jar that will spawn, colonize, and fruit?

Because it is MS you cannot guarantee much.. it is a crapshoot every time.  Also, every time you plunge a needle into that LC you create a vector for contamination, so you cannot even ensure it stays clean.  That said, I've used them with decent success. 

I am hopefully stopping from doing many more MS grows by cloning a good cluster on agar and making a LC out of that, or just doing the G2G like our friends above mentioned.

Oh, and no spore print/syringe is 100% clean.
That's another reason we don't recommend inoculating LCs with MS.

Right. I have a colonized rye jar from an LC. I haven't fruited it, but could I hypothetically do G2G with this jar as the master and have viable spawn?

I would suggest you fruit first and then isolate or else all you will be doing is using a multispore as a master, and you will most likely have inconsistent flushes from that.

Just because it colonizes doesn't mean that it will fruit successfully. Even if it does, which I hope it will, you will not have the amazing flushes that you see some of the users here have done. Most of those have isolated a successfully fruit, by cloning, and made masters from that.
