What's going on with my agar cultures?

Hello mycophiles,

I recently inoculated some fresh petris with fragments of rhizomorphic mycelium from a P1, first generation (spores), petri dish. It seemed like a very healthy sector. All petris were inoculated from the same P1 dish.

Does anyone know why the first petris show a somewhat darker growth spot in the center changing to a whiter, more rhizomorphic growth on the edges? Could it be that the mycelium was still recuperating from the shock and then got healthier from a certain point on?

In the 1st and 4th pictures a triangular spot of almost no growth appears, why does this happen, anyone knows? Maybe there is some whitish mold I haven't distinguished...

On pictures 5, 6 & 7 a cottony growth appears. I'm not sure if this looks like cubensis mycelium. On picture 7, 2 types of growth appear. Healthy rhyzomorphic, and this cottony type. Could it be a kind of white mold, or is it a differentiated sector of cubensis mycelium?

I'm no expert but it looks like you are growing some green mold in those.

I'd toss em.

You don't toss petri dishes when contamination shows up.  You transfer clean mycelium to a fresh dish, and then repeat the process until you have a pure culture.

The darker color in the middle is normal.  It's also normal for mycelium to change from cottony to rhizomorphic and back.

That dark green in the middle is normal??

I'll take your word for it because I've never tried agar, that just surprises me.

Correct me if I'm wrong, but isn't that dish beyond saving or would you actually attempt to cut out some of that rhizo?

You again with a satisfying reply, thanks a lot. I really didn't think they were contaminated with mold because I have encountered molds that are much greener, but I was somewhat concerned. I trusted the fact that normally molds outgrow cubensis mycelium in no time and inhibit its growth, and that my plates show rhizomorphic growth up to the edges; but with your response now I am very confident that my plates are mostly clean of contaminants.

Good to know that it can become cottony back from rhizomorphic. Thanks everyone for your replies.

If I really had to do a transfer, it would be from the third pic.

The rest look like way too much work.
