Spotting Sectors and Isolation Questions [PICS]

Recently a friend has begun attempts to isolate a strain, but I don't have enough experience with this to offer any advice. I'm wondering if any of you see any sectors on this dish:





Those pictures are the same dish from different angles.  My thought was that maybe the mycellium just hasn't colonized enough of the surface of the agar to see sectors yet; is there usually a certain diameter at which sectors become more apparent?  The other reasons I've come up with as to why either of us can't spot any sectors are a) lack of knowledge regarding what sectors look like or b) perhaps this is already an "isolated" strain (I'm still not sure I fully understand the how exactly the genetics work when isolating a strain); the dish was inoculated via a single piece of colonized bird seed which had been inoculated with a spore syringe.

My understanding is that, assuming you're trying to improve yields, it would probably be best to have a clone rather than an isolate because the clone is surely going to fruit, or at least has a better chance than an isolate (just my understanding so far).  So, my question is this: assume a clean piece of tissue from a fruit body is allowed to colonize a dish of agar; will that be an isolated strain, or will it produce sectors, and if it produces sectors, would there be any benefit to isolating this further?  Or would that be relatively the same as starting from a multispore inoculation, then isolating?

I couldn't find "isolation" or any version of the word in GGMM, and I've done my best to search for the information I'm looking for, so I apologize if this has been answered to death.  I appreciate any time taken to help us out.  :grin:

Not in my experience.  Not seeing any for sure in those pics either, except at 6o'clock in the first one and you won't want that likely...

Any and none of the above, depending on the circumstances of germination, colonization, and territory disputes between more aggressive strains.  Except that, yes, if it sectors, by isolating further you'll end up with a predictable strain, although it may only be predictably poor performing.  That's why you want to test all the isolates thru fruiting.  YMMV of course.

Chances are yes, it will sector.

Yes, there are benefits to isolating further.

Thanks for the straightforward answers guys.  My understanding is that this particular plate ended up with a little contamination around the edge (perhaps that's why the 6 o'clock point in the first picture doesn't seem to be colonizing?), and a small piece was transferred to a clean agar dish.  Hopefully I'll have some more pics next time I see my friend.
