Growing from a print... Have a few Questions.

Okay, so I have like 20 good BLACK Spore prints that I got from some cinctulus. I also still have the dried mushrooms.

I really want to grow some mushrooms already!!! I live in California so I can't order and spore syringes, and California isn't really the best hunting place either :sad: However, I found what the good people of Shroomery confirmed to be Subbs; Took good prints of them, and fully dried and stored the shrooms. I have read and read and read about how to grow out of popcorn, rice flour, grain, horse shit, cow shit, and everything in between... I JUST WANT TO START MY GROW! The problem is I don't have agar as of right now, and still live with my parents so getting supplies is a little hard. Will someone please offer some advice on what I could possibly do to make this happen or am I just at a loss until I can get a spore syringe...???

in the top right corner is a search box.  type in "making spore syringe" and start looking at the threads that come up

you're not going to want to make a syringe from wild prints, it's likely they also have traces of other mold, fungus, or bacteria which will lead to contamination.

Agar work is necessary, if you dont have access to it - wait until you do


also, be very selective with your prints - some of your pics looked like there are a few foe's mixxed in

Are your parents cool with you growing mushrooms? If not, you really shouldn't be growing them at their house anyway.

Well anyyyyyways. I got my hands on some agar powder. I also have some petrie dishes, sterile hay meant for infusion kits, and I have packaged sterile soil...  I can grow outside or inside, but I'd prefer outside. Plus I have unlimited access to cow, or horse dung. My backyard is grass, and it's shaded... I know the general different ways I can go at this, but are there any tips you guys can offer me, seeing as how I'm in So cal and it's so damn hot!
Please, any advice you have! I will do anything to grow, I've researched, and hunted for about a year now and just want to start already. If theres anything completely necessary I'm missing I will find a way to get it... haha  :sporedrop:  :sporedrop:

you should also pick up a pressure canner/cooker (PC), make a Still air box (SAB), raid your USPS or FedEx location for tyvek envelopes, and ditch the petris, get a pack of Ball elite 1/2pint wide mouth jars instead

This is obvious.

Materials:
1. Distilled Water or Tap water that had been boiled for at least 10 minutes.

2. Syringe

3. Spore Print

Steps:
  • Get a sterile area (Preferably a glove-box or a Hood).
  • Scrape the Spores into the water (That is sterile and cooled down).
  • Suck the water into the syringe and press out again into the container where the water was as to get all the spores off the sides.
  • Done, you have a syringe.

yes thats how you would make a nice n dirty syringe:thumbup:
wild prints are never clean neither is the mushroom, inside and out so agar is is the more logical route.
welcome to shroomery 2 btwsdanky(and i don't bother chasing spores off the sides, once it hit's my syringe it doesn't come out till inoculation, spores are microscopic so if you can see it, thats alot

Just prepped my agar with Potato Dextrose (Yeast-extract) Agar (PD(Y)A)... Plus I went out yesterday to my horse stables, and found a few fresh samples to clone. I didn't feel to confident about how old those prints were. Plus I think the old "BLACK" prints were just wishful thinking they're kind of like a black brown. I figured it was better safe than sorry, the new prints are BLACK I put a dot of sharpie next to them and it matches!!(: Yay. So time to start.  :dancer:  :rockon:

Should I use the base of the stem, or the gills??? And how long do I let the agar cool before pouring into the dishes? Then how long do I wait to actually place the spores/fruit in the petrie dish?  Sorry thats the part I'm super confused on...

agar turns solid  at like 105 F keep it just above to pour and your good, when its solid its pretty much good to go :thumbup:\\

to clone take a tissue sample closest to the bottom of the stem, I would think if you tried to clone from the gills you'd risk germinating spores at the same time kind've sabotaging your clone :shrug:

Make a syringe from a wild contaminated print
use only a few drops from syringe for agar and return to storage
bacteria continues to reproduce while in storage
now you have a shit syringe full of bacteria

Not saying it's not possible, just that there's methods for better results



you're better off just scraping spores onto agar in a SAB

for short term results it doesn't really matter though :shrug:

yeah you can clean them up, but the syringe is likely to go to shit making it useless in a few weeks

I hate syringes so as soon as I realized you were discouraging their use I'm like "why am I disagreeing with this?"

Really all that's holding me back from PE is my distrust of anything in a liquid suspension and swabs are hard to find :tongue:

I'm a bit drunk to boot :shrug:

after pissing away 2 syringes i made from some giant mutant fruits, i'll never look back.


I finally got results dropping the veil remnants of one of those fruits onto agar, and now it looks like i may loose that grain spawn to bacteria  :ifyoucanawe:

And swabs usually don't get handled/sterilized/dried properly...every single swab I've gotten from someone else has been covered in bacteria. However, the few I've made myself worked like a champ :shrug:

I don't like spore syringes either but if you want a PE syringe, go get one from a sponsor, it's totally worth it :thumbup:

Im really tempted but, I seem to have rotten luck with syringes even from sponsors :shrug: of all the syringes I've gotten from sponsors, only one worked, most grew nothing and I had a one beautiful specimen of aspergillus. Really most of my successful grows have been from prints acquired from the good people in the marketplace :super:

Maybe i will take a chance and try once more for some PE or APE before i retire :wink:

It sounds like a bad thing but I'm not bitter or nothing. If anything my poor luck with syringes only made me determined to learn to use agar and grains. Who knows I might have been content to dick around with PF tek a lot longer otherwise. So from that point of view it was probably a good thing in the long run.

I started using agar heavily not too long ago for cloning and don't plan to turn back. It's simply too damn easy. I feel bad for the dozen LCs that will probably never have a use.

I think I just started to get the tiniest little bit of growth on a couple plates! White fuzz, Yay!(: hahaha  :whoo:  I really hope they don't get contaminated, what are some good ways I can prevent this??? Also, should I be putting water in these? They seem dry.....

No the agar will provide the water. Excess water is an invitation for bacteria. That's why when using spore syringe to agar you only use a single drop and try to not let it roll around on the plate.

see the top left? that's contamination, what you would want to transfer is healthy mycelial growth as seen at the other locations - but let it grow out at least 5-10 days before transferring to new media. You can continue to transfer as long as you like, but once you have clean growth it's ready for grains or your choice of proper media.

VERY healthy growth on one plate! YAY.  :girlishjoy:  :ohboycomputer:  Fairly good growth on a second, the others don't really seem to be taking. I varied the parts of the mushroom I used, Spores, base, gill ect. The two plates using the base of the mushroom are DEFINITELY better!!! I am soooooo excited... now how long do I let this grow before transferring to either a grain or bulk substrate of wood chips...?
I can do either, any advice on this decision as well???
I'm kind of looking for quantity over quality, for the first time anyways :P ... I just want to grow them already!!!

PS: No contams on these two plates are seen, should I still do a transfer???

I found them on Grass???? I just didn't think simply grass would be a substantial substrate...
I'm sorry, I've read many threads about transferring mycelia to wood chips, and that just came out as an example...
I know they like stable-like areas, would it work if I transfer to grass with aged/ partially broken down horse manure with some hay or something like that??? I would just prefer to grow out side if possible...

for outdoors a mix of h-poo and grass clippings would be great!i'd start piling up the spots right now and turning the pile every day till it's dry and aged, you don't want green grass clippings in there. i would love to see some pics of your dishes

Its all destroyed!!! God damned contaminations!  :friendlyfuckyou:  Screw thissssss. Hahaha I swear, I'm going to cry. Mushrooms are the so important in my life! But, idk how long I can afford to keep buying $20 an 1/8. I've never been so excited about anything in my whole lifeeee, now what do I have? A total failure...  I transferred and then left them for a day, and the agar plates dried out! Why do the mushroom Gods despise me?!?! Will someone please tell me a BEGINNER method I can use... My biggest problem is no one will deliver spores to Cali... And I think I'm using contaminated prints.  :icecreamsadness:     
I dont know what I can do, I want to just buy everything I need and do it 100% "by the book" so to speak, for my first time... buttttt whats the point of spending that money if I'm not even gonna be growing anything thats worth it you know? haha

you should watch the agar section (maybe all of it and even buy it - I did :shrug:) . the musics a little rough but That will help you learn how to "clean up" a spore print. basically once you see growth you transfer the growrth to a new clean plate - isolating it from contams. sometimes you only need to do it once, sometimes three, somtimes you don't need to at all.

I'm sure some kind gentlemen of the shroomery will be willing to send some cubensis prints, to an attractive young cali girl like yourself.

(cubensis are a more beginner friendly strain)

They teach agar work to high school students nowadays, so it doesn't really get any more noob-friendly.  And seriously, don't fret.  You have several things going for you:  you live in a mountain of h-poo with pans growing all around you.  Any mistreatment you receive here that's not due to the internet being the internet is probably jealousy based.  You know how many people here would kill for that setup?  Virginia Gentleman is one of them.  Seriously, don't give him your address.

You wasted thirty cents worth of agar on those plates.  This is not a life-ruining failure.  Get on your horse and ride around until you see more clonable specimens.  Make like ten more plates at the same time, and do it with more water this time.  You're shooting for 2-3% agar by weight here.  That's 3-4.5 grams per ten dishes and 150mL water.

If you want to splurge, go get some fish antibiotics for like ten bucks at the pet store.  Tetracycline is broad-spectrum and will definitely help with bacterial contamination.  If not, just transfer away from contams onto plates you already know to be good (because you were able to grow your clone on one made in the same batch).

It's a basic idea of mixxing and sterilizing agar, then introducing samples and waiting

wash
rinse
and repeat until you have clean growth


not many people are willing to help if you do not have the incentive to learn on your own. it's simple

Whoa guys, take it easy.  Shroomery is supposed to be about community and encouragement (and it usually is), but seeing disparaging threads like this makes me question whether or not that's the case...Messing up sucks bad enough without being insulted for it.

That being said, don't get discouraged. Failures are the best experience if you can tough it out. 

I know you can't just order spores, and that sucks.  I'd personally do everything you can do make Franks Cloning Tek work if somebody doesn't come through with a syringe or print.  If you can get some clean myc on agar, there is no reason that growing it out to finished boomers should be too much trouble.  Read and read through everything FrankHorrigan has put together, and then read it again while you put it into action.  He has done a stellar job of putting together reliable methods.  The best shortcut is to follow the teks exactly,as any substitution or straying will welcome problems.  If you read Frank teks closely enough, you won't even have to ask another question to successfully grow.  He's seriously put together a step-by-step guide for growing start to finish.  I like tmethyl and Notahacker420's tek collections as well.  If you are really careful, you can seriously just follow along without knowing much theory; you'll certainly gather a fair amount just by doing EXACTLY what they say to do.  Oh yeah, you will get mushrooms too!
