Tyvek on PF jars.
    #1826147 - 08/18/03 12:41 PM (20 years, 7 months ago)

I have some pf jars, and innoculated 4 days ago. I used tyvek filters on the lids(1 layer only). I have yet to see any growth. I am probably just being impatient, but will tyvek give enough air for a pf jar. BTW, i didn't use a top verm barrier, due to lack of spare verm. help me please!

4 days is nothing, write back in two weeks.

OK, a few of things...

You say you used tyvek and no top verm layer.  How did you cover the holes in the lid and tyvek after you inoculated? Or did you just leave the holes open.  Tyvek will effectively block contamination, but if you leave the needle hole through which you inoculated open, a vector can sneak in.

Did you shake the syringe before inoculating?  How much solution did you use and at how many inoculation points?

It can easily take a week for growth to show up and if your temperature is much below 85F, it can take even longer.

Finally, it's INOCULATION, with one 'N'.  Get that straight!

Danknugget that's more than enough air exchange. I wouldn't be surprised if your jars dry out actually. Through trial and error my friend it's a waste of time, in his experience. He has a bunch of tyvek but just uses tinfoil now with no air exchange during colonization, still gets cakes done in 2 weeks time.

Either that or your jars dried out due to the tyvek.

i used tyvek on pf and popcorn, and i did not inoculate thru the tyvek, slightly lifted it under an oven sucker vent, and I think the popcorn looks pretty dry, so i squirted some sterile water in one of the popcorn jars, to see if it would help. i put that jar in my computer case to incubate it, and left the other ones in tupperware near a sunny window, it should get a bit hotter there. Thanks for the help!
