Question about negative pressure while making honey water...
    #1827770 - 08/18/03 09:18 PM (13 years, 5 months ago)

The mole on my left testicle was making some honey water and has some concerns regarding contamination. The jars (4 at 125ml each) , and water and honey were sterilized via boiling (2 for 20 minutes and 2 for 30). I then waited a day to inject the spore solution. Upon removing the tape (there was only one hole in the lid for an injection site) 3 of the jars "sucked" in air. I carried on with the preocedure (which was all done amidst the updraft of my oven) and put the jars in a warm place. So here's my question: from peoples friends' experience, is it quite probable that the "sucking" in of air from the jars caused contamination?

If so, is there anything i can do to prevent contamination?? maybe incubate at high temps, add h2o2 and hope the good guys hatched and survived in time and the bad guys die off? also...really, what's the chances of honey water contaminating? honey should never spoil...

Thanks for any help. My nut is just looking to confirm or toss out his worst fears.

couple of things your mole could do on the next go .
cover the jar with foil, and wait till jar is cooled before he tapes the hole .
or, draw sterile air through a flame with a syringe and inject through the tape .
sense he used the oven tec (and if he did that properly) probably nothing to worry about .
tape over the inoculation hole isn't realy needed unless you have a problem with ants, like i did .
