Workman, what is your opinion on inbreeding?

Inbreeding?  Well, it isn't generally good long term, but it is a useful tool for enhancing and isolating certain traits.

Each multispore culture is selfing (breeding with one's self).  This doesn't seem to be a problem initially, but sequential multispore cultures reduces heterozygosity.  What I mean by sequential multispore is using prints from your current cultivation to start the next cultivation and so on.  A better idea is to save prints from your earliest cultivation and use those to preserve your "strain" as long as possible.  Cubensis spores should last at least 10+ years if kept cool and dry, but viability drops yearly, so you may need to use large amounts of spores to revive a culture from very old spores.

The loss of heterozygosity means that the spores will produce fruits that show less and less variability from multispore, which sounds fine.  You get a nice uniform crop with little variability, so its similar in behavior to a clone.  But this also makes your culture vulnerable to random mutations.  Random mutations that can be recessive and invisible to the grower.  The PE mutation is a good example.

The PE mutation is recessive, so lets say you made a multispore culture from a print, maybe you selected out a single strain on agar or just injected tons of spores into a substrate.  You got a crop of nice looking mushrooms and printed some.  You gave away all the prints you made and just have one print remaining.  Unknown to you, the mushroom you saved the print from was generated by a pair of spores, where one had been hit by a cosmic ray that damaged a key gene necessary for proper cap development.  The mushroom looks fine because each of it's cells also contains the nuclei from the other spore with an undamaged gene.

You only have the one print, you want to grow it out again and expect the same results as before.  But something is wrong, several mushrooms look like PE, with weird malformed caps.  The selfing, or inbreeding has resulted in pairs of the damaged gene appearing in 25% of the generated strains.  Many of the mushrooms in the crop look fine, but most of them will also contain the damaged gene.  And this is just one trait.  More mutations can sneak in over time, almost all of them are bad, some can make the mycelium unable to even fruit.

So save your earliest prints, save slants of your best cultures.  Combining the spores of different varieties can increase heterozygosity which can then be selectively selfed to produce a new custom made variety.

I know it's confusing and I probably didn't explain it very well, but maybe I helped.

i got some spores from an old hippie who said he had been growing them for years and taking his own prints, after a sad performance he tells me that he might be over using his variety, meaning growing/printing/growing/printing/growing/printing and tells me that over time, the result of this can be weakened genetics and it might be time for a fresh start.
is there any truth to this or am i hanging out with a man whos done too much acid?

so maybe he hasn't done enough acid!

That was explained very well. And with simple enough terminology that the average Joe could understand. Thank you.

I think the last couple of pages have been some of the most interesting posts this thread has ever had.

QFT.  I'd love to work with those PE looking fruits.

Here's some more Redboy pics from the recent past.

Straight horse manure , no verm. First flush. All ropey and longer than I had ever seen them by far. Many were close to 16 inches. All had veils that broke and fully opening caps.

The stalks were very white , a trait of this Redboy.

Even though they had nice veils and fully opening caps , many had distorted gills. They still printed nice and dark though.

That's the second flush of the same tub and you can see that this time there are many mario-like fruits in the population alongside the open-cappers.

Some dark blueing going on.

Here's same spores , same manure , in a different setup. I grew a food container of manure/smoved it a yogurt container with verm on the bottom, that I bottom-dunk throughout the grow. All opening caps on this one.

Same thing but fruited invitro in 2 litre pop bottles with verm at the bottom for bottom-dunking.

After the ropey long stalked , open-capped flush in the straight manure tub came the march of the Marios. They were the majority of this flush.

Close up of a nice Mario growing.

Marios on the same sub as in the bottles , manure pressed into a cup , colonized and then flipped out of the cup. I do that sometimes in bottles , but also in tubs like cakes , but I place each one in the watering tray with verm.

Mario cluster.

Big ugly Super-Mario , and friends.

Chubby , thick Marios.

This is the 'Medusa head grow from in the ziplock bag harvested. It was about 250 grams wet or so. All ropey , opening caps , nary a mario.

Gills from the fully opeing cappers in the first flush , straight manure. They still give decent prints.

Redboy mycelium growing through straight horse manure

Sorry about the randomness of the presentation. Just going through recent pictures.

I actually really like the openness of this Redboy. It has a wide gene-pool of specimins. I find the potency through the roof. You have to be careful giving them to friends because they will ask for them by name for years and years and it will build unwanted notoriety.

Nope.

I just have an appreciation for good explanations.

The ratio of shitty one line responses, vs legitimate explanations is pretty thin on this site. Workman explained the process, and it was a good, and understandable explanation.

I'm currently of the opinion that, "UTSE" is about the worst response that can be given. Better to cite the source, or a good example.

Hello everyone I have a question and I'm guessing this is the place I'm suppose to ask it.

So I've been doing some thinking and searching and I was wondering about this problem with headaches that I have with Golden Teacher when I take two grams and above. It's the only cube I've tried so far. Now I searched on shroomery for "golden teacher headaches" and found a few threads with people who's used Golden Teacher's and have gotten headaches the day after.

I have been thinking about trying Cambodians because I've actually seen one or two threads where people said they didn't receive headaches the day after, but I truly want to try South African Transkei because I've heard nothing but great things about it, it sounds like it can be perfect but I never could find any talk about the day after feelings of them.

So I'm really just wondering from people's experience with the cubes in question have anyone ever received headaches on high doses of Cambodian's or Transkei's? I know this sounds crazy to some because I read all over that "a cube is a cube" well I'm not asking about potency or how the trip may differ, that's irrelevant.

All I want to know about is the after effects the next day, I really love the way mushrooms feel but I at least want to consume an 1/8th and every single time so far with GT they've given me unforgivable headaches that won't allow me to go that far again and one gram doses just leaves too much to desire for. Last time I tripped it was on 2.5 grams and it was fine but still leaves a desire for more and the headache happens still so it's not worth it. Regardless if people think it's pointless or not I'm going to try one of them to see the results I get anyways because I really have no other choice, but I just thought I'd ask to receive more insight on this. I'm just trying to find out as much as I can about this before just completely giving up.
