i have  used fsre already and got some cubensis spores and yes i used a 2 Euro coin ;D
this time i want to try and grow shiitake

and gonna try it on a supermarket dried shiitake coming in a plastic bag hopefully itll work

Do you have any idea why shiitake would so rarely produce a fruiting substrain? You would think that evolution would naturally have encouraged the species to frequently develop fruiting strains.

if im not mistaken i saw the answer to this question and some1 said he saw RR replying this be4 and the answer was because shiitake got very aggresive mycilium, so if it already started growing somewhere it will continue like"forever" or something.. again im not sure:D

Shiitake is just picky.  I've grown from spores and managed to get mediocre performance, but that was after a very long time trying.  This is the reason shiitake is traded in culture form.  Somebody else has gone to the trouble of isolating a good fruiting strain from spores.

It's the same with most species actually, including the cubensis many people around here are fond of.  Multispore inoculation is a roll of the dice.  Sometimes you get the jackpot, other times you have three mushrooms on a whole substrate.

Dry mushrooms can sometimes be cloned.  200 years ago, all spawn was dried for storage and shipment.  You can also place a piece of gill tissue on agar and grab spores from that, but you'll definitely have to isolate any mycelium from all the contaminants that are going to appear.

This probably warrants a thread unto itself, as I'd like to pick up on what RR's just mentioned. I had assumed that, as is the case with cubes, putting shiitake spores on agar and isolating an apparently vigorous sub-strain sector would yield something very likely to fruit well. With cubensis it's normally the ropey, rhizomorphic sectors that tend to produce fruit; but by the sound if it, it isn't so easy with shiitake as simply looking for the most promising sector, or else we could all do it at home.

So how do they do it in the labs that produce the cultures we buy? Do they use a microscope to be sure that both + and - nuclei are present in the isolation, and thus able to sexually bond as is required for fruiting? (if I'm not mistaken, when only + or - nuclei, but not both, are present in the protoplasm that mycelium fails to fruit.) This discussion makes it sound like when shiitake spores germinate, they are more likely to grow without fusing with neighboring mycelia to share nuclei, or that if that happens, then the + and - nuclei are for some reason reluctant to bond and give rise to a fruiting sub-strain. Or perhaps with shiitake either the + or - is rare. If it's true that fruiting sub-strains are hard to produce, there must be some reason.

If the culture-producing labs are not using microscopy to isolate good fruiting strains, then how are they doing it? Because if a culture has to actually fruit to prove itself a strain, then strain isolation is a labour-intensive exercise that carries the culture a long way from the spore before it is selected and offered up for sale.

A second related question is about the cultures sold; at FP, for example, you can buy shiitake in various forms, the test-tube culture being the most expensive. I had thought about buying a bag of plugs and simply putting a few on agar, but would this be giving myself tissue of a much older generation? Maybe it's worth the extra price for the test tube culture.

And RR, when you buy a culture in the mail, do you replicate and preserve G2 cultures so that you are you good for the year, or do you frequently order fresh cultures?

Thanks for taking my questions, which like mycelium, branch off in all directions... (aren't you guys glad I limit my time on shroomery?!)

I only order cultures once.  My shiitake 75 culture is nearly ten years old and still performs as well as the day it arrived. I cloned my oyster strain from a wild fruiting in 2004 and it's still in wide circulation, being sold my many vendors.  If properly cared for and preserved, you can keep young cell lines for decades.

People who isolate good shiitake strains do so just as any other species.  You isolate dozens, or even hundreds of strains until you find a good performer.  There is no need to use a scope other than to check for contaminants.

When you receive a slant, inoculate a petri dish and then make a dozen new slants within a week as soon as the mycelium starts to grow.  Don't let it fill the petri dish before grabbing a new culture.  As soon as you know it's contaminant free, make new slants.  Save the original.

It's a good think you didn't try to come up here Friday.  We got 18" of snow.  You'd still be here.  Hopefully, the snow plow will make it up by Monday or Tuesday, 'cause the stores are begging for shiitake and I still can't get out to deliver. :frown:

Very doubtful.  If you want to grow shiitake, pony up the dough for a quality culture.  Why start with a lead and mercury contaminated specimen from china that is only in your country to ensure the local farms go bankrupt? Remember, 80% of the shiitake sold world wide are from china.  Aside from the fact that normal working people can't compete from a business standpoint with a communist system, you'd be starting with a senescence ravished culture, dangerously contaminated with heavy metals.  Why bother. :shrug:

Unlikely but not impossible. An interesting challenge for somebody with skillz and time.
If you can get a fresh mushroom, you could clone it on some agar. That would be way easier.

Without stipulating to that, may I ask if you have numbers for US-sold shiitake?
Because it would not surprise me to learn that Japan, for example, imports mushrooms from China.
But the fresh (not dried) mushrooms sold around here are mostly grown locally.

It's somewhere around 60% of the 'fresh' mushrooms sold in the U.S. that are from china, and around 80% of the dried.  You can find the latest numbers with an internet search.  With the several hundred pounds of shiitake that Mrs Rabbit and I sell weekly, there's no way we could hire an employee, even at minimum wage to work for us and beat their price.  Workers in china average around $200 month in pay, but farm workers make much less.  In the U.S., we can't even buy a weeks worth of groceries for that.

Yeah, I don't know what scale you'd need to be on to hire a crew, but it's big.
I know you wouldn't hire illegals, or legal folks illegally, either.

Canned mushrooms might be from China. Not that it's relevant to the thread.
I'm going out today, I'm going to check some local stores for fresh and dry mushroom sources.
Not exactly doubting the info, but I just don't recall seeing mushrooms grown anywhere but here.
But I'll check it out. :thumbup:

In the next county over from ours, the supermarket, had Canadian white button mushrooms labeled as 'Product of USA' until I threatened to expose their deception to the FDA.  I had asked them three times to observe the law to no avail.  The boxes the mushrooms came in were clearly labeled from Canada, but still they kept the USA label and american flaga on the compartment in the store until I pitched a bitch.

This was the store that refused to buy my product for no other reason than it was produced in the USA.  They said after the contaminated peanut butter, green onions, and broccoli scandals they would not have any more American produce unless everyone had a million dollar insurance policy, a requirement they don't impose on imports.

Here in Canada I've grown frustrated with stores who just plain don't seem to know where their shiitake come from (or so they say). So I have to assume they're from China. I also found tomatoes labeled BC Hothouse but with stickers from Mexico (which I boycott because they affect endangered peyote habitat). I have a pretty deep mistrust for the big grocer chains, and none of them ever agreed to carry my packages of wild-picked dry mushrooms ("needs to go through head office", they'd say). Free trade be damned, we need to legislate that a percentage of products sold be local.

But back to topic; this thread has me thinking that I could use my leftover dried morels to make spawn, something that I hadn't considered before. The dust at the bottom of the bags must be full of spores.

RR, I'm only about 60 miles from there as the crow flies, but these last few days here it's been one 4-inch dusting followed by nothing but sun. Yes, great advice you gave me about holding off, as I'd be snowed in till who knows when, trying to earn my keep as your unpaid alien workforce.

i get your point but the problem is im from israel and its literally
almost impossible to import a liquid culture even with standart air mail, it requires an approval of the ministry of trade and Agriculture
and a realising fine , ofcourse they dont always catch the goods but lets put it this way:sadly theyre pretty much efficiant. only way to go is to use spore prints(which wont work in the shiitake case) and the other way to clone a supermarket dry fruit.
these are all the ways i know of, hopefully some1 will enlighten me
