Amp resistance, why wouldn't this work?
    #1811433 - 08/13/03 09:21 PM (20 years, 8 months ago)

I work in a developmental genetics lab, and I was thinking about the issue of contamination, and I think that I may have a simple solution. I can easily transfect an ampacillin resistance gene into the spores right next to a omniexpressive gene. All you'd have to do is add some ampacillin (a strong antibiotic) to the growing medium, thus eliminating 99.999999% of contamination. This is really easy to do too, wouldn't take longer than a few hours.

Did you figure this out with Ohms law ?

ok...well if it works...send me some of those spores...could you bioengineer p. cubensis to be more potent as well??

ummmmm. what!?

I think it's a joke relating to the title of this thread, "Amp", which the originator meant as an abbreviation to the cell-wall antibiotic ampicillin. The reference to Ohm's Law interpreted "Amp" as the abbreviation to Ampere and made fun of the word "resistance" which is measured in Ohms.

By the way, anyone know why abbreviation is such a long word?

I don't quite understand.

Ampicillin resistance is inherent in fungi already, as it acts on [many] prokaryotic's biochemistry.  This would reduce it's usefulness in combating mold contamination if the transfection were successful.

Ampicillin plates will germinate spores just fine.  It's actually my antibiotic of choice because it's so cheap and available.

As far as 'easily transfecting' next to a normally active promoter, well. . .I guess we have different views of what is 'easy'.

I think Suntzu has a point... and hell, if you can use your lab for genetically modifying physcadelic mushroom spores, can't you just grow your mushrooms there? That would be a very easy way to combat contamination

how would you know this gene your'e transfecting would be expressed?? wouldn't it have to be within the omniprescent gene you referred to?? if so, what vector would you use to get it there???

i'm intrigued.... keep us informed
