Invitro Pins not growing in FC, no new pins developing (2.5 weeks in SGFC)
    #18025841 - 03/28/13 08:22 PM (10 years, 10 months ago)

Hello, my first round of cakes I left a little too long in their jars, and they started invitro pinning like mad. The biggest grew to the size of a small shroom, and pushed it's cap above the dry vermiculite barrier, maybe i'll get some photos up later if anyone's interested.

All this started happening during a very busy time of my life, I had no time or materials to build a fruiting chamber, so by the time I got around to it, all but one cake had multiple invitro pins.

Now, since they were growing so well inside the jar, I figured they'd do better in a proper SGFC, but it seems they were growing faster while they were in the jars. Since i've birthed the cakes, there has been very little growth, and only the smaller pins are growing (however I did notice many of the pins that grew upside down immediately began to point their caps to the light, not much growth after this though).

I have a SGFC set up in my closet, there's about 2 inches of space between the FC and the closet walls on the short side and about 5 inches space between the long sides. After reading up on misting & fanning I figured I was misting too much (5 - 6 times a day), so today I've cut it down to 3 times a day but i've continued fanning 5 - 6 times a day.

The temps in my room rarely drop below 65F, and I do believe I have good humidity in my SGFC. The perlite is slightly moist to the touch.

One more thing, I cased my cakes with some verm on top.

So how come the cakes aren't developing new pins, and how come the invitro pins aren't growing? They've been in the FC for about a week and a half, do I just need to wait longer?

I dunked two for 16 hours and two others for 12 hours, some pins came off during this and the birthing. I've heard that longer than 12 hours is detrimental and can set the mycelium back, could this be the reason?

The first (and biggest) invitro 'pin' when I first noticed it:

A little bit after:

That particular mushroom hasn't grown at all since I've began fruiting it, and it looks quite scraggly.

Wait longer..... yes

Less misting.....  yes

Fanning....you are doing enough

Other than that you look good to go to me.  I've had some strains take as long as 11 days to even start thinking about pinning.

Got it, i'll wait a while longer, in the meantime i've set up another round of jars, cooling down tonight, inoculating tomorrow.

It's been about a week, here's an update:
5/6 jars I inoculated show signs of germination and mycellium growth. The four cakes currently fruiting appear to have done nothing, no pin formation, the invitro pins haven't moved a bit.

I'm having the same issue.

At first, my SGFC wasn't proper so I fixed it to fit RR's construction.

Next, I cut down on the misting.

The Cakes colonized the verm roll, and I'm now on day 10 mwith absolutely NO PINS and NOT LOOKING GOOD.

I have been so patient throughout this process, but this is really pissing me off.

I wish I could figure it out, but I cant, and I'm now too nervous to put my other cakes inside this SGFC - though it is correct.

Good luck - I wish I could give some advice, but I just wanted to let you know that you are NOT ALONE.

One big thing I feel i've missed in this thread; the cakes were in the jars for a lot longer than they needed to be (hence the amount and size of invitro pins). I birthed them ~5 months after inoculating, 3-4 months after they were 100%. Is this what I could attribute the problem to?

5 MONTHS?????????

I can't imagine that being beneficial - the myc is probably WAY TOO exhausted at this point, if it wasn't put into refrigeration/dormancy.

Just toss them and start over, and try not to get too discouraged.

Better luck next time.

Sounds like they were way to wet or battling a contam or something to take that long. I had some GT's take almost a month and a half to get to 100% visual colonization - Still waiting to see how those will fruit.

Long story short those are past their prime :/

do WBS next time, if possible.

However, lie everyone else, you should try to get at least one semi-successful PF out of the way before advancing - it shows you all of your mistakes and forces you to adapt to the fungi.

Good luck, though... you will have success if you keep trying.

The jars didn't take that long to reach 100%, there were also no yellow metabolites, if that's any indication of contamination. I also think my sterile procedure is decent, out of 10 jars I haven't had one contam (yet).

So I should toss these cakes? They're not doing anything still, unfortunately. No way to salvage them or anything, or are they spent?
