dudeman has a problem with his jars.
    #1805615 - 08/12/03 02:49 PM (13 years, 9 months ago)

2weeks post inoc. no signs of anything but some slimey lookin stuff in 4 jars. just a little water stain brown crud(isnt growing...looks like it might have been too wet)he thought he had growth before...he was wrong.

he has 12penis envy and 12 malabar pints with wbs. polyfil in holes. temps are 73-85(non ac'd room)

he put .6 - .8cc/pint then shook immediatly after inoc. (hes thinking that was bad advice) its also very hot where he lives and he thinks it might have been hot enough to kill spores during delivery.

should he....
1. use left over spore solution to REinoculate 2 of each and try for a g2g trans?
2. get NEW wbs and inoc 2 each and try g2g from that
3. go new wbs AND new solution(cut losses and start over)

HELP!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!

1.No if th jars never had any growth for 2 weeks DO NOT re-inoculate them you may just waste the last of your spores.(make some new jars up because it sound like you have too much water in the jars and the mycelium won't grow in that invroment but contams love it)

2.if you do this you will have to make up all new jars

3.I would go with #3 " go new wbs AND new solution(cut losses and start over)"

dump all the contents out of the jars and make new jars but use less water this time(if you used too much last time) make new jar with new substrate in them then take the last of you spore and inoculate the jars if you want you can inoculate 2 jars then do g2g to the other jats that have been sterilized

Spores straight from syringes in WBS jars may take a 4 days/week to show signs of growth. This works fine, but it allows too much drying and/or contam problems so a certain percentage of jars can easily be lost.

If you have a bit more patience and make PF cakes in jars then use the crumbled core of each cake to inoculate the wbs jars you can be more certain of quick growth and ensure less time for problems to develop. It's not likely the best and certaintly not the only way, but it does work.

Of your three options, option 3 is best. Cut your losses and try again, because trying to rescue those old jars is pretty certain to fail.

Huh? Why would you wait for a pf jar to colonize just to use it to innoculate some WBS? If WBS is that hard, use popcorn. It is easy!

2 weeks later isn't too much the problem(it could get too dry) but the too much water you say you have in the jars is a very big porblem as you already found out

alright what should the consistancy of the wbs be like?

it looks plump mostly...and upon turning over will easily fall around jar. wbs does not stick together.

popcorn tek anyone?

dudeman's method of preparing birdseed:

take penningtons wbs with red, white millet, sunflower...and some little grey specks looks like kitty litter.

empty whole bag in big bowl. put water over seed till its standing 3/4"

let sit for 8hrs with aluminum foil wrapped over.

unwrap stinky seed. drain and rinse(really not much draining going on) but he rinsed real well.

fill wbs into pint jars 3/4 full. add lid with 1/4" hole filled with polyfil(he has tyvek now and will be using it if he can find support for it.

wrap tops of jars with foil. pc for 1 hr.

remove from pc and shake. none really looked "dry" all looked hot and wet.

waited a day. inoculated in clean room using modified oven tek(mod: i forgot to turn it on, just kinda inoculated in the oven)

shook directly after inoc.

waited and waited...

what sounds like the problem? THANK YOU FOR ALL THE RELPYS SO FAR!!!!!

i've seen different methods for preparing wbs. one is you let the wbs soak in water for 24 hours then rinse well and fill your jars. the other is to rinse the wbs really well then fill the jars with the wbs and some water. they both involve rinsing the crap off the wbs, such as keote and stuff.

in your preparation it sounds that you let it soak for only 8 hours, and didn't add any water before PC'ing to compensate. maybe there is not enough moisture in your jars?

""maybe there is not enough moisture in your jars?""
see that is a concern too.

how can i tell "proper" moisture content of jars? all i can find are pictures...they "look" like they should be good2go.

the white millet is hard and shiney the red millit is plump and the shell is translucent(not transparent, nor opaque)

the "water stain" i refered to earlier looks like.....the seed was wet and was giving off some water onto the area(1mm)of jar it was touching. just little watery lookin spots.

he also added several glasses of water to the pc before he cooked em.

he wanted me to tell you also that there was a black rag(cotton, old, clean) that he put in the bottom of the pc'er to keep the jars off the bottom. the water turned black in the pc'er.

could THAT have any effect?

no, dyes coming out of the water shouldn't affect it as it has happened to a friend before. i don't know a good way to tell if the water content is to high or low, besides the obvious pooling of too much water. give it more time, or make up some new jars.

a friend recently inoculated 12 pints of wbs to find only 9 colonized after two weeks. he didn't clean the remaining 3 out or anything just left them sitting there on the shelf, and lo and behold one of them started colonizing more than a month after inoc.
