Germinating Panaeolus Species

I am currently studying evolutionary characteristics of certain fungal clades. We are trying to germinate Panaeolus cyanescens and Panaeolus cambodginiensis spores on MEA plates, but have not had much luck so far. And the spores do come from a trusted Shroomery vendor.

We first had them sitting at room temperature, but moved them to an incubator at 25.5 degrees Celsius after two weeks. It has been approximately two more weeks since we moved them into the incubator and still no luck.

Does anyone have any suggestions on how to help germinate? I have a whole lab of equipment to use, so if there is anything else that might help it along please let me know.

Hmm, I think your syringes were not loaded with spores, or you're not agitating the syringe before inoculation.
Perhaps even the agar was prepared incorrectly?

I've worked with 5 Paneaolus species, some wild spores, and some bought, and they all germinate very quickly and colonize
rapidly on all types of agar.

Germination times in all my experiments were within 5 days. Typically the 4th day.
A temp of 80F I found to be optimal for Paneaolus.

I wish that were the case, however, I can actually see the spores on the plates under the microscope, so I don't think not having enough is the issue.

As for the plates, I've germinated various other genera just fine. Maybe Panaeolus prefers something other than MEA? Though it seems to be working well for everything else.

I've germinated the spores on PDA, MEA/LME/ELME and one other I can't remember the name of, with no issues, I'm curious to find out whats causing the issue here, just as much as you are. :lol:

I guess the best thing to do at this point is simply switch to a different agar and see what happens.

I've had a similar experience with straight MEA, change your media around or supplement it a little, see what happens.

Obtain new spore print your wasting media.

Viable spores from those species germinate readily on malt extract agar.

A record of the date when a spore print was made is useful.

Hi Chrontaunomo, I have recently germinated Pan Cambo on MEA. It was also taking awhile for me. It took like 9 days before I saw any growth. I would just try making another plate. Also you can try if you like, but I don't know if I would work towards an isolate. I would try to make transfers for thicker growth. I had to make a transfer or 2 because my initial mycelium was rather whispy. As soon as I got some thicker growth I waited till I had about a quarter sized piece. I dropped it in the jar let it colonize a bit and then shook. I have shook twice now and my jar is fully colonized which I intend to g2g. All the stuff you hear about "dont shake Pan jars" I say don't worry. I have been shaking with no issues whatsoever, infact they only look fucking stronger after I shake them lol :shrug: Just reattempt it on another plate, I have faith that you'll get it. Shake the syringe! Every so often I completely forget too!

i tried a pan cambo once and had no problems. :shrug: i never had the time to do anything with it though.
there is definetly harder shit to germinate then pans!! try wild Agaricus!!:lol:

Please explain.

BTW
Panaeolus spores germinate readily.  You do not need any special media. You need viable spores. The advice your getting about changing the media is foolish. Get fresh spores.
