When to shake WBS jar. - MS -
    #17923877 - 03/08/13 11:31 AM (10 years, 11 months ago)

So I knocked up a dozen or so jars of WBS just over a week ago. As far as I've read, when you have a MS jar, you only shake it at about 30%. Problem is when I knocked them up, I just let the spore solution drip down the middle. Now a large majority of them are showing me growth. Some little spots at the bottom, some at the side of the jar. Which meaning the myc in the middle is growing and expanding.....Soooo, since I didnt inoculate the jars with a method that would easily show me growth, how do I know when they;re about 30%, so I can shake em up?

It honestly doesn't matter. You can shake once, twice, three times, whatever you need to speed along colonization.
Shake now and see what happens in a few days time.

Ah, see I was under the impression that shaking only prolongs colonization. I mean I've seem the arguments about it, and shaking MS vs shaking wedges, but I thought the end thought was it was bad.....Well...good to know, time to shake them fuckers
:crazy2:

I'd just let it go until you can guesstimate it around 50% and then give it a shake to make sure the colonized grain is evenly distributed, by the time it recovers it should only be a day or two away from full consolidation.

People say it prolongs colonization if you shake right after you inoculate. But shaking after it partially colonized speeds it up because you are distributing that myc all around the jar.
Citric is a big fan of shaking his jars ALOT. Or I should say was a big fan. I think he said he had fully colonized MS quarts in 6-8 days? Somewhere close to that.

You don't need to shake it a lot when using a liquid culture or doing a grain to grain transfer, depending on the amount of inoculate used the average for a qt/L 8-10 days for full colonization/consolidation.

I made a 2Gallon W.B.S Grain spawn master, used 10cc's of spore solution and shook only once around 45-50% so, roughly around Day 6, by day 12 it was recovered fully and 14 consolidated.

If its a agar wedge, then 2-3 shakes would be required.

Too bad Citric hasn't been around lately.
He'd be here telling you about how he colonizes a jar from MS in a week by shaking frequently :awesomenod:

I found this post about Citric's method for shaking. He also incubates his jars so that speeds things along a bit.

Quote:

Citric said:
If people would just give it an attempt at proper incubation they would see increased colonization times.

I shake when I inoculate, regardless of MS, LC, G2G.
The day after first signs of growth, I shake.  The first signs of growth if I shook well enough after inoculation they would be roughly 25% colonized.
Depending on how they recover, I sometimes shake the day after recovery.  If they recover from the initial shake with inoculation points every half inch, then I let them go.  Just need to shake the hell out of them.  When they recover they are usually 50-75% colonized.  Two days later they are usually 100%

80-83 degree TiT.

I broke both hands and had surgery on my right hand and have two pins through my knuckles to my wrist.  I also lost the internet and found out my girl was cheating on me on Valentine's Day, so life been a little hectic!

But yes I shook at inoc, day after first signs of growth, and depending on how they recovered I would shake again the day after recovery if they were not spread well.

Hah, thanks.  Yes it is challenging.  As bad as it sounds I am use to it as I break my hands often due to being a professional boxer.

I'm over it all, shit happens.  Keep it moving
