Pc petri dishes?

Hey all, I would like to start working with agar, so i went out and got a pressure cooker todat after reading that steam sterilizing isn't recommended for agar. I have polystyrene petri dishes. Can I just pour my agar in them and pc them? Or do i need to pc my agar before I pour it? Thanks for your help i have a lot to learn.

PC your agar and then pour the dishes in a SAB or in front of a Flow Hood.

I fill glass plates with unsterilised agar and PC them all the time. Plastic plates melt.

....technically, different agar formulations require different amounts of pc times, depending on the nutrient base SOme as few as 20 minutes, but i just do them all for 45 minutes (16 psi as i am at 3000 ft) to be safe. make sure you cool them down before pour; i bring mine down to 55 degrees celcius.

I would be afraid the plastic dishes would warp in the boiling water, dunno, never boiled plastic petris before 
I think they're UV sterilized before you purchase them; if you're thinking about re-using plastic ones, I would be apprehensive about contamination (even if I had some UV sterilizing abilities at home) personally, I just bite the bullet and order new ones

sterile polystyrene petridishes are usually sterilized by gamma rays after packaging. they are generally intended for single use only. once unsterile, there is no way for you to get polystyrene sterile again, so work wisely.

Gamma rays are great, because they go through everything. For about $3 million, you can have your own gamma ray sterilizer. Not counting the cost of the cobalt-60.

it was a cover up. he was really trying to re-use his agar dishes. the initial frustration of having to buy more made him turn into a big half deaf guy. sad day for science...

Your mileage may vary...but I have had very good success with soaking pre-used petris in bleach water until right before pouring the agar. I also did some experiments with 2oz and 3/4oz deli cups...no contams. Obviously I got lucky with that.

Bleach does not kill all mold spores, neither does H2o2, neither does alcohol.
But heat does.

Man, I'll never touch plastic plates again. Grrr. Glass it is. Just sent an order to my favorite supplyer. I can toast them until every contamintant turns into charcoal.

What about lysol they claim to kill 99.9 percent of viruses and bacteria.I also just searched lysol and they claim to kill 99.9 of molds and fungi. Just a thought but maybe hold the can upside down spray all of the air out then puncture it pour the contents into a bowl to soak your petri's in. I still would suggest investing in Pyrex if you want to use them multiple times. They can be found for around $5 a piece.

goddam Pyrex! 5 bucks a piece, you can use them multiple times, slippery when wet, heavy, makes you feel like a scientist, cry when you drop one...plastic ones are dirt cheap, light weight and disposable...

Yeah I have dropped one it sucks. There is a place here in town that sells them for $16 dollars a piece lol. I wont ever buy one of theirs they have probably had the same ones for years.

Ive had much success from using salsa jars as petri dishes. They wont boil over while in the PC like petri dishes do. You can also put a silicone injection port in if you like. I dont use anything for gas exchange as i dont store my plates for long.

Peracetic acid-based sterilizing and disinfecting solution provides high-level disinfection and is completely effective against spores. Ready-to-use solution is tuberculocidal, bactericidal, virucidal, and fungicidal. Completely biodegradable, it decomposes into oxygen, water, and acetic acid.
I know its used in the food industry.
I wouldn't use it to disinfect dishes before use. But I think it would be good to spray down the room and work surfaces.

I will also put dishes with agar in the pc I do not have aproblem with them boiling over though. Maybe that could be from over filling them or releasing the steam too fast. I do this because I only make a couple at a time.

I realize there are more then one way to skin a cat. I may have over reacted. But I was taught to use a flask or bottle for the agar and to pour the hot liquid into sterile dishes in front of a hood.

if you PC petri dishes with agar in it, it's important to wait for the pressure cooker to reach normal pressure before opening it, otherwise the dishes will boil over. i allways PC my petri dishes prefilled with agar, but for more than a decade i'm just working with glass dishes. they cost about $1.50 in Europe. started with a package of 10 - meanwhile over the years i'm working with hundrets of them. i try to avoid plastics where i can - have you ever thought about the ecological consequences of discarding hundrets of plastics petri dishes in a year or even a few month?! glass petri dishes can be cleaned and used over and over again. initial higher cost but on the long term it is cheaper for me to work with glass.
