Encouraging rhizomorphic growth on plates
    #17832808 - 02/19/13 09:05 PM (11 years, 2 months ago)

I have used plates off and on, but never had success in seeing rhizomorphic growth or significant sectoring. I don't know if its the media I use. I have tried spores as well as cloning.

My media typically includes maltose and yeast extract. I use the same for LC and have great growth.

Why do you want rhizomorphic growth on plates? More vigorous growth (if that's even what this form means, which I'm not sure of) on agar doesn't necessarily translate to the same behavior in another medium.

As for sectoring, how many transfers are you making? It can take a number of transfers to narrow down the genetics enough that sectoring is even possible.

I want rhizomorphic sectors so that I can identify isolates that may perform better. This is pretty standard for working with agar.

You can use slightly less nutrients in the agar, this should make the rhizomorphic/more aggressive mycelium stand out, and kind of run ahead of the rest, in search of more food.
