Saving Mycelum.
    #1776543 - 08/03/03 10:12 PM (13 years, 4 months ago)

Here's a situation my friend from holland was explaining to me, he'd like an answer as soon as possible.

his jars was innoculated with very old spores, maybe 3 years old. Anyway, of the 5, 2 became contminated with orange/rust contam both at 1 innoculation spot. The other 3 just plain didnt grow anything.

Bieng lazy, he left the 5 jars in the incubator another week, in case something grew in the 3. Over the weekend, he came back to find thier was no growth on the 3, but 1 of the jars that was contaminated shows sign's of mycelial growth 1 hole away. The orange did not grow over the weekend. Thier is a fairly good distance between the growth and the contam (see pic).

this is the last bit of his syringe and print. And the strain could possibly be lost if he does not save it.

The question is: what can he do to save it. He was thinking of doing some liquid substrate and transfering some of the good mycelium into the jar. He has some food grade agar (almond flavor).

also, just to add, that this is C.Tropicallis, but that shouldnt matter much (except to deter the people that say "just buy a new syringe of teh strain" since it's a bit more expensive than cubensis)

A quick reply would be very helpfull. Thanks.

i would do at least 3 or 4 jars, or agar plats, for better chances of success .
add 1 ml 3% h2o2 to 100 ml liquid or agar .
i have no experience with flavored agar, but i haven't heard that it wont work .
good luck .

i'd recommend using plates rather than liquid. and definately do a few, as deanofmean said. i have saved strains like this before...

good luck!!!

but would food agar work? The ingredients are

sugar, Non-Dairy creamer, Agar Agar e406, Silicon dioxide 551 (anti caking agent) and artificial flavor.

my buddy told me that the sugar looks pretty dominant. 130g for 3 cups of water, which would mean somewhere around 110g of sugar&Creamer.
