Agar time out of the fridge before inoculation
    #17420969 - 12/20/12 08:04 AM (11 years, 4 months ago)

I started using agar a few months ago and really enjoy it however lately I've been having some issues with it. My cultures seem to go pretty well and once they are nearing the edge of the dish I put them in the fridge. This didn't seem to cause any problems for me until recently. I think I may not be giving the mycelium enough time outside the refrigerator to recover before inoculating.

How long (minimum) will you let the petri dish sit outside the refrigerator before you use it for inoculations?

A day.  You might want to put them in with a good half inch form the edge to avoid contamination, then when you take them out, wait until you see visible growth.

There is absolutely no need or reason to warm up cultures before transfer.  Simply make the transfer and return the master to refrigeration.  The transferred culture can recover in the grains as easily as on the petri dish.

makes no difference.  I take them straight out of the fridge and make brownies just about every day.  myc doesnt care.  and i never incubate it.  my lab sits at about 60 degrees so i am always favoring myc over bacterial contam.  I have decided that slow and steady growth is the way to go on agar.
