If you sterilize the the HBWR first then, used distilled water in a half pint and pressure cook, then transfer HBWR to water in front of flow hood or GB or whatever. Contams shouldnt be a problem, if you got the seeds make several jars, trial and error, all you need is one to work then you in the clear.

I have some fluorescein. Used in cultures with Claviceps to identify larger producing sectors. Can this be adapted for our needs here?
I would b happy to send a little bit out if it will help. What I am looking for now is Ehrlichs ... anyone?.....

At the moment, we really need to just isolate the varying species present in/on the seeds and leaves. From there, identifying with microscopy against the published information will be valuable for eliminating growth of undesired isolates. At the point of specific, desired isolations sector production analysis may be a useful test...

Since the seeds have the desired fungi growing epiphytically rather than endophytically might it work to simply sterilize the seeds and place them in the agar medium whole without crushing in order to prevent contamination by the unwanted endophytic fungi?

If done right sterilization will kill all bacteria and fungi inside and out of the seed. Sterilization would probably render the whole process obsolete when it comes to seeds. Best bet is to try to isolate on agar with multiple transfers, jus know what your looking for and wht it ooks like so your not making a beefed up contam

Hi guys. In case my absence has not tipped you off, I have been derailed from this project permanently. Hopefully I inspired you good people to try some science out; with luck someone will continue this research. I did find the following article, which came out this year.

It outlines some good phylogeny of our fungus, but classifies Periglandula as "un-culturable". I think we will see more references this coming year.
