Blue Meanie (Cube) Isolates?

I was hoping I could get some input on if these all look like isolates or if I have another transfer to do. My next set of plates are ready for transfers if it needs it...otherwise into the grain! (and agar slants)

They look real nice and thick! i bet they would produce very strong mycelium growth by the looks of those. get um onto some grain and test to see which fruits best!
im working with the same species as you! i placed one grain colonized rye grain per plate, and did three transfer generations after that. In the end i found 3 different growth patterns. Now (not pictured) the transfers from these potential plates are looking very "mono" like and thick like yours.
Plates i selected to do a Fourth and final transfer
    
and after three transfers from the original grain plate these looked promising to me. I transfered two chunks from each dish (not all pictured) and saved the original dishes in the fridge (these pictured plates).

The bottom right on 2b looks the best IMO. If you take a tiny bit from there I bet you'll have a good isolate. It's hard to tell which is best though, you should decide a little earlier next time. It will be easier to see before they've hit the sides.
How many transfers is 2b from the original MS culture?

I believe this is after the the 3rd, maybe fourth transfer (I really need to either slow down my smoking or start keeping records)

Beautiful. I've always wanted to work with cubes... they seem so fun and easy to work with. Here's a couple tips that might help:
1.) don't let it grow out to the sides of the dish/over the sides of the dish 2.) first MS/clone should be marked on the plate as M1/C1 or something of that nature, and the following plates should be T2 (2nd transfers), T3 (3rd transfers), etc.
I did that with my first try at agar (learned it from reading MycoElf's posts), and I wrote down the strain, date, and M1 or T2 etc after every plates. It was extremely organized, and very helpful for me as a beginner.
that also gave me an opportunity to stack them on one end of my flowhood one at a time, then sterilize my scalpel and sanitize my gloves before the next go
these should turn out beautiful, huh?

Yes I am hoping they do turn out beautiful. I wasn't too worried about being super organized about the transfers since it doesn't really matter except for having the knowledge of how many transfers it took to isolate.
I have marked each section of each plate (as Top, Left, Right) and will keep perfect track now that I'm at the isolate stage.
I'm the same way with the Psilocybe Galindoi and Psilocybe Tampanesis agar plates as well

Well whenever you get the master grains going... post a link on here to the cult forum so that we can all see how this turns out! Definitely nocc up at least 10 master slants too so you can preserve those genetics!

10? master slants? There are 18 isolates....10's not enough. but yea I plan on making slants prob sometime this weekend
