a few questions from a novice

hi all, ive been cultivating magic mushies for over a year now and have previously had some awesome harvests with casings and a marthas closet i was using. it pushed out some good weight but moved to monotubs and im having problems, im certain i am not having contam issues here but here is my problem:

I have cased up in monotubs (using montubs for the first time btw) using the 10:10:1 verm, peat moss, calcium carbonate casing method. my monotubs are plastic storage containers about 10" wide x 13" long and 11" tall. i have two 1.5" diameter hole cut on two side at the top and bottom of each conatiner stuffed lightly with polyfil. the casing are about one month old now and have just started to see any real fruiting. pinning seemed delayed compared to past operations . and now it seems i have tons of aborts. i fanned the tubs at least four times a day and mist the same. i became concerned about not seeing pinning so to increase fae i hooked up an aquarium air pump run through a hepa disk flowing to each container. i know it wont make tons of difference but i was hoping for something. my casing became fully colonized before ever fruiting so of course i now have more side wall growth than anything. along with very heavy bruising on top (again 100% not contams). I am very disappointed with this first small flush and am hoping for some answers or recommendations as to what i can do to improve. temp hovers around 74-77F and humidity in the tubs is right at 90% at all times. what am i doing wrong?? thanks for any help!

Ok, im still a newb but troll this place frequently. Did you do multispore, or isolate? The concensus here is "multispore is a crapshoot." So every batch will exhibit differing characteristics, including colonization speed, immunity, potency, fruit bodies, etc....

well im not sure what those terms specifically mean, isolate vs multispore. i colonized in whole grain rye berry jars using PE syringes. all current fruits seem to be the same strain. not any varying characteristics except size.

You will want to do some research on how to isolate on agar. When you use a spore syringe its referred to as multispore. With literally between thousands and millions of spores in your jar many of them germinate, and produce a monokaryotic mycelia. Monokaryotic mycelia dont fruit. They need to find a mate. Which is another monokaryotic. When it does it forms a dikaryotic culture, with unique genetics. Theyse dikaryotic cultures vie for control of the jar. They all have differing characteristics. They are all related, but more like cousins than brothers.

When you isolate, you pick just one of those dikaryotes with the features you like, and experience fast colonization, good immunity, consistant flushes, and fruits.

Cool thanks for the knowledge! Its odd I've never had this problem before with previous batches, they all seemed to pin and be very uniform with fruiting. I've previously looked into agar but u seemed to solidify that its gonna be the most efficient route. I also homebrew so ill be able to use the agar culturing for both hobbies, myco cultures and yeast cultures. whats your opinion on the best method? cloning or isolating? it seems either would be better than MS.

For starters, im just regurgitating. I havnt produced a flush yet. Your ahead of me there. Im also a brewer. I thought you just used some dme and some water with a breathable plug for yeast cultures. Cloning is a form of isolating, so you should be good there.

agar would be for storing yeast cultures for later yeast starters in brewing. you can use tube slants or petri dish. slants store for longer periods. i would use liquid culturing for both hobbies, with a erlenmeyer  flask and stir plate.

Thats a great plan. I usually just buy the same yeast, but have used the previous batches yeast as long as my timing was right.

ya repitching is my prefered method but timing is rarely right for me. i do change yeast often too so even if timing works its not always same yeast.

Well from what Ive trolled around here... Did you let your substrate get too dry? Did you touch it where its bluing? Did you use any chemicals in  your monotub that may have damaged the substrate? How is your air exchange and moisture levels?

This is where the more experienced folk need to chime in.

used no chems. never touched substrate. it may have dried out but ive been misting every day so i dont see why it would dry out. air exchange seems good, i fan multiple times each day and i have the aquarium air pump hepa filtered going to each tub (5 total) humidity maintains around 90%.
