Some tips for PF Nobie's

Nothing ever goes quite as one would expect, and when one actually starts unexpected things tend to unexpectedly happen.

Id thought making the syringe itself would be the hardest part, and yet it was inoculation where the difficulties really began!

Many things didnt quite pan out as expected, but the biggest problem I hadn't anticipated was the needle getting clogged in the brf and verm substrate.

Have since found this is fairly common frustration, but at the time I couldn't really understand what was happening, so I simply pushed harder! Boom!

For a flustered frustrated noob fumbling and flailing around in a GB this was a real nightmare and bred further mistakes such as pulling air and verm (2nd to last jar) into the syringe but mainly as stated massively over shooting!

Had 12 cakes and due to blocks and over shooting had six still left to knock up after using up 15mls worth of solution! And that was by no means after having managed to get all 4 holes on every jar!

2nd time was an improvement with 7/8mls knocking up the remaining 6 jars, but overshooting was still a big problem (didnt get all the holes on every jar!) though this time I sucked air into the syringe from within the flame of my alcohol lamp or simply burnt out the verm!

Another problem I found was with the micro-pore tape! I realize now when using this one should use tiny pieces that simply mark out where the hole itself is! Course you could also make the holes bigger than 1.5mm!

I myself spent ages tapping the needle into the plastic lids before eventually finding the hole, it was very fiddly and unsettling and not nearly as smooth as I had hoped for beforehand.

A learning experience for sure, but I think grain jars with one injection port have to be so much more easier than farting around with all those holes, not to mention making the cakes themselves.

Have no idea what the success of the mission will be but Im not exactly optimistic! Colonization times from jar to jar is likely to be all over the place as well, even if it works! I just at this stage cant really envision anything happening at all to be honest!

Hopefully this will help my fellow noobs be aware of a problem that I wasn't (there are posts about this but no mention in the main teks I think).

Anyway if you get blocked dont panic and push harder, but remove the needle, burn out the verm, and try again. If that doesn't help you can suck some air into the syringe but as I learned do so from within a lit flame to prevent contams!

Hell better yet practice with a syringe full of water and a tub full of verm, might sound silly but I wish now that I had!

I'm still not exactly sure how to overcome this problem entirely, its really not good to be going in and out the GB all the time and for a noob especially it adds to a mounting sense that everything is going wrong.

In all likelihood for me in future I will just avoid the pf tek!

All in all a bit of a disaster!

It sounds like you used a too-small needle.  I use 14 gauge needles which never clog.  When you wipe down the tops of your jars with alcohol prior to inoculation, push hard and this puts a small divot in the micropore tape where your inoculation holes are, making them easy to hit on the first try.

Actually I didnt know I was supposed to wipe down the tubs with alcohol first! I thought the foil would have kept them clean,another problem there perhaps!

Many of the tubs did indeed have dents in them (I had worried over this problem before hand) though some didnt and it was these exceptions that caused the problems!

The needle seems pretty big already I thought it would be better to buy a thinner one to be honest giving that less verm would be able to get in?  I see so thicker will make the injection itself more forceful pushing out and away any blockages?

Anyway not the end of the world have many prints to practice with and it was a good introduction on the whole!

It can only get easier now!

I had a similar experience...(minus the clogged needle drama) and everything turned out fine. I got 0 contams and my time cleaning myself and my environment proved worthy. Yes it can be stressful as hell the first time... but hey, isn't it always :shocked:
