Which creates better grows? better spawn, or culture??

So I'm mid way through my first ever successful bulk grow!!! It's going REALLY well, but for my next grow I would like to attemp to change one factor at a time so I really get to understand what I'm doing. My question; which would show me better growth (speed, size, potency, and uniformity) making a better spawn recipe (I currently use damions50/50 coir, but would like to use like 30/30/30/10 hpoo/hay/coir/Verm) or doing culture work and isolating a strain? I'm guessing isolation is the answer, but can it seems a lot harder then making a stronger bulk substrate. Also, do I have to pastureize? Or can I just stick it all in a 1 gallon glass jar and pc for 90 min???

I would culture tissue from a fruit.

You need to pasteurize bulk subs. Verm/coir is forgiving but I'm not sure about PCing. Also, use straw rather than hay.

Isolate monocultures on agar, maybe from a fruit body, then fruit each one out to see its potential. Keep the one you like, ditch the rest.

As long as you are using MS inoculations, your speed, size, potency, and uniformity will vary.

Straw in substrates can be difficult I hear/read, but RR has a video on how to prep it if you are interested.

If you PC your substrate, you will contaminate. Pasteurization is required for manure substrates at 140-160F for 60-90 minutes. Here is RR's jar pasteurization method. :thumbup: There's some others in my sig links as well as other good info. on bulk growing.

PC is used for grain/wbs preparation at 15psi for 60-90 minutes and any agar you prep to clone, sector, or isolate at 15psi for 30-45 minutes.

For the best growth, it makes sense to me to have a balanced diet (for us and our mushies :mushroom2::hug::amanita2:). Check out The Additives Thread from CH HELL for good nutrients. You should get gypsum in your mix, and I really like coffee in mine too. I'm also looking into alfalfa and kelp meals. My current standard substrate recipe is horse manure, coir, verm, coffee, gypsum at 50/20/20/10 + gypsum.
