Very interesting that you rate micropore above tyvek, I've seen a lot of people stating it's totally useless! I've started using it for a half-assed GE port on my LCs (double layer) so am happy that's not the case

I generally use sfd's but when i run out I use 2 layers of micropore tape. Not sure why people would say it sucks, cuz i've never had a single contam in a jar using micropore tape. The only contams ive had were using polyfil and using a tyvek paint suit.

Had better luck with envelopes than with the suits everyone says are better.

Just my experience.

Oh forgot to mention I typically see faster colonization times using 2 layers of micropore than I do with sfd's. Can't explain it.

Has your experience been with MS or with isolated monocultures?

with both actually. ms I typically just put 1 layer for the pc and then noc with the syringe and place another over the first. with agar i just use it as a filter and it works just fine.

Got any pics of your isolates on agar?

I'm a fan of rhizos!

Im planning on doing grains. Isn't the 4 hole GE for BRF jars?

If I cut up the tyvek in quarter sizes how can I "double up"?

By placing a layer on top and the bottom of the lid.

There are no rules, I'm not talking about 4 holes  at the edges a la pf tek, I'm talking about 4 or 5 virtually touching like:

x x
x x

In the middle of the lid, in order to stop wet grains from directly touching the tyvek.

And yep, top and bottom or just stick 2 layers together. As I said, that seems too messy for me so I've moved to polyfil directly on the lid and tyvek elastic banded over the top.

Me too, I just knocked up 24 master jars with PE isolates I've been working on for a couple months.  Average of six transfers to monoculture.
Agar is great.
